I'm now on the path for killer genetics.
I think after I spawn these 18 jars, I'd like to take a step back and work on something fantastic before I make more tubs. LilOldMe has great tubs, and pasty among others say it's the genetics, not how he's running it. Which I agree with.
I can make agar. I can do transfers etc.
Now how do I get killer genes? Slants? Keep transferring a thousand times? What's the method to the madness.

Tons of transferring and testing is the only way to find great genetics once you get a killer tub slant it and either keep on transferring to get better results or just work with that slant ... lil has a bad ass gt culture but you can have killer tubs with giving the best enviorment for your mushies

Cloning is a good short cut. If you start from spores and just isolate. You will have to do tons of test grows some of which might not even be fruiting strains. With a clone at least you have some idea what to expect.

Oh shit, welcome back! Sup with isolates? I've read you say that you recommend going back to spores several times a year and I'm curious to know the reasoning behind that. Why would you do that if you have a killer isolate? You can keep a culture for a lot of years without it losing strength, can't you?

I keep a chunk of myc on the pasty plate that I inoculated that killer tub with?
That's there I'm not grasping. Keep a thousand pasty plates in the fridge that I'm testing, and the best one, transfer that myc and make a slant??

In nature they don't have refrigerators to put them into periods of slow cell division preventing the shortening of the telomeres. Keep your masters young, rotate their media and they will last a long time.

So you guys are saying you can only expect different results from cloning a fruit from an MS grow? Trying to clone a nice fruitbody from an isolate is pointless?

Yup. Once you've isolated down to a monoculture. There's only one strain. The genetics can't be narrowed any further.

You slant a good isolate or clone. If you have a monoculture that you want to test, transfer from the donor plate to more plates, keep one on standby in the fridge and use the other/s to noc some grains to see how it performs. Same with clones. If it performs well, take the culture from the fridge and slant it.
