So i posted some pictures of a possible blue bruising pluteus sp recently. I didnt get much in the way of an ID. I decided to do a spore swab and tissue clone. I have growth from the tissue and the spores. The plate with spores is heavily contaminated. The tissue not so much.
My questions are
1-can i put a piece of cardboard in with the tissue? If so how wet should it be? I read that i should get it wet then squeeze it until no water drips out. Is this accurate?
2-What would be the best way to attempt to clean the spore plate? Normally with cubes id toss any contaminated plates. I read somewhere to pour hot agar over it and wait for the mycelium to grow through to the top and transfer from there. Is there a better way or should i just do tranfers until its clean?
I havnt worked with woodlovers so im kind of in over my head. Normally i do a lot of researh before attempting anything. This was a spur of the moment thing.

Cardboard sounds pretty useless if you already have it on agar. As for the swab, was the cotton swab sterilized beforehand? Give it time and you'll have cube mycelium escaping the bacterial pool. May need a few transfers before you have a clean isolate.

Sorry if i was unclear. Its not a cube. Its what i believe to be a pluteus sp.
Also, sterile swabs wouldnt really help since it was done on a wild specimen. Though it was sterile right out of the pack.
Its not growing very strong on MEA but thats all i had on hand. Thats why i was asking about the cardboard. It is a woodlover after all.

Even woodlovers thrive on agar. They can break down cellulose and sometimes lignin and eat that, but agar is like a milkshake to them. If you're having slow growth, it's probably just because of the growth rate of the species, or the temperature of your place, or maybe a hidden contam. I wouldn't put cardboard in there.

Yes, do a couple transfers to make sure your culture is clean and then you can go to spawn, just like with any other species. After you have your spawn, though, woodlovers get tricky. For some wood-loving species people use a small amount of grains to inoculate pasteurized sawdust to make colonized blocks and fruit those, like a shiitake. I'd try that first, but there are a lot of other options like inoculating whole logs with plug spawn or making a wood chip bed.

I love those spawn plug logs! It's a shame the shiitakes take a year, but I find that whole process to be super enriching, and if I ever have the natural resources on hand...,I'd try my hand a year long shiitake grow!
