Contamination OR Mycelium?
    #23489027 - 07/29/16 11:26 AM (7 years, 5 months ago)

Hello! I want to know why the two plates pictured (F2 and F3) display two colony formations. It appears as though there is one central colony with a smaller one trying to escape, for lack of a better term. Can anyone tell me if both colonies are fungi mycelium OR is there some contamination here? If they are both in fact mycelium colonies what could explain the double colony formation? This is my second round of plating in my attempt at isolating a rhizomorphic monoculture. I started these from an original plate that I inoculated with a spore scraping. This is my first attempt ever at working with any kind of culturing techniques .

Hard to tell it looks fine. I would avoid that area in your transfers.

Agar questions go in regular old mushroom cultivation by the way.

Last link in my signature has all the up to date agar threads pictures and videos

Rhizomorphic has nothing to do with monoculture or even performance.
Using less nutrition in your agar makes mycelium usually more rhizomorphic and using nutritious agar tends to more tomentose growth. You don't know the performance by appearance. Healthy growth is all you need to be concerned with.

You will have to test the cultures anyway. I would say you're quite lucky if 1 in 10 monoculture is even worth fruiting. They usually don't perform nearly as well as a clone culture would.

Thank you for the valuable info. Basically, I am wasting my time when I can just make a tissue culture from a hearty fruit. Is that sort of what you're saying?

Also, this was my fist post so next time I will consider more thoroughly where to post.

Yep a tissue culture is a way better option if you're looking to grow something good right away. Do the isolating in spare time and hopefully land an awesome isolate eventually
