Three Super Quick Questions

It's been a while, so I want to come back properly with style.

These are my three questions.

1- Can I pressure cook Pasty plates with my grain jars for 90 minutes? Will the extra time ruin them? I'm wondering just for convenience's sake.

2- I really don't have much to elevate my quart jars (glass ones), is it fine to have them at the bottom and putting water up to the filling line in my Presto 23 quart?

3- I used to use Tyvek for filters but I've had no luck finding them recently. I bought polyfill instead, never been really good with it though. SHould I make patches by ironing them down? Or just do the big hole and stuff it through?

No, the pasty plates will be fine.

I only elevate my jars using the little tray that come with the unit.

I just popped a piece of polyfill into the lid and sterilized it with foil over it. I wanted to blame it for my contaminates, but I think I was just using the wrong seed mix.

I wouldn't run liquid media or agar for longer than about 50 min at 15 psi. You run a serious risk of caramelizing the media which will result in sluggish growth.

Yamoon you have been kinda shitting the bed with questionable advice today.

I use ceramic to keep the glass from touching any aluminum surfaces.


Two pieces of micropore tape in a cross pattern over GE holes has worked for me.  Can do a 3rd in asterisk pattern if you really are worried about contams floating in.

I agree, tyvek is easier to work with than polyfill. You can find the paint suits for cheap on ebay or if you live in the US, you can basically find them in most hardware stores I hear.

Ironing poly to make a felt like material is good too but the comments I read about it make it seem like you need to get used to doing it a bit, it might take a few tries to get it right. Felt is said to be as good as SFDs. I wouldn't use polyfill as a filter personally, I hate it.

I also agree with pasty, 2hrs is wayy too much for agar.

I mean, who am I to argue with the creator of pasty plates? I just mention what works for me. Haven't had caramelized petri as of yet. May differ using potato dextrose instead of malt?

If your in the US you can have free Tyvek sent to your house from the USPS or pay 10$ for better Tyvek at most hardware stores or maybe even a paint store. You can find Tyvek here in the sticks

Seeing this disagreement beetween two growers I respect gave me a reason to look this up.  It turns out MEA does in fact caramelize more easily than potato dextrose, possibly because malt extract contains about 1-2% fructose.

Caramelization points for some sugars:

Fructose 110° C
Galactose 160° C
Glucose 160° C
Maltose 180° C
Saccharose 160° C

So Pasty's recipe, which calls for HFCS, will caramelize in a pressure cooker more easily than potato dextrose, which is pretty much synonymous with glucose and won't easily caramelize in a PC.

Looks like there's room here for everyone to be 100% right and strictly talking from experience.

Glucose, fructose + vit C + H20 + long boil = invert sugar.  Which never crystallizes either.


For non-grainy fudge etc  :laugh:

I actually use MEA myself :shrug:
Just worded it weird because of a wake and bake.
I also tend to let me pasty plates cool off in the PC.
The sudden temperature change would otherwise make them a bit caramelized on the bottom of the plate. The tops always work fine though. I have heard that I poor a bit thicker than necessary too though. :shrug:

Haha oops definitely went ahead and pressure cooked the plates for 90 minutes before others commented, I guess I'll find out how that went. they seem fine? I used spores on them so that I can hopefully get a nice clean culture, accidentally way too much on two of them, but whatever, learning process.

I have a question on storage for the pasty plates now though.

I have one plate leftover for hopefully a transfer from the best of the 3 I inoculated.

Should that guy go in the fridge to last longer? The inoculated ones just go in the storage area correct? Because the fridge slows down the growth.

don't pigeon hole yourself into one recipe.
You're not KFC.
The myc likes to eat something else once in a while. Keep your cultures virile; change the agar recipe around.

No, store unused plates in room temp, you only store plates in the fridge when you have a culture you want to preserve. A plate will stay good for months given that it stays clean.
