Agar is antibiotic agar from here http://www.fungi.com/search-results.html#/?q=Agar
And for the Petri dishes i make my own no stir with this containers from big lots Use food coloring in the agar it really helps define contaminants and how well the mycelium has grown

Melt a 1/16 hole in top and cover with micropore tape an pc for 45 minutes

Do you PC for 45 minutes at 15 psi? Also do you get your micropore tape from Fungi.com as well? Do you just use 1-2 drops of food coloring per dish? Also why do you have all your dishes tied off in the clear plastic bag?
On the website for the agar it states, "50 grams are recommended per liter of water." So you are mixing this with water before placing it into the containers correct?
Sorry I am a complete newbie at agar if it isn't apparent.

If I'm not mistaken, it's normal for the stipe of PE to look this way. Maybe I have my strains or mutations mixed up. Correct me if I'm wrong. I've been sucking information here like a $10 whore and may have a contam.

I think he meant 45 min @15psi. Yes You can get 3m paper tape at cvs or Walgreens. On the back of the package, top left it will say something like: in the medical world this tape is known as micropore tape. If you notice there is condensation on the inside of the tied off plastic bag, I think he pced the "Tupperware and then placed them in the plastic to cool and store til ready. I don't know the answer to how many drops he used.

Yes to all your questions that's exactly what I do the bag around the agar are just mushroom bags I filled in pressure cooked to save for a long time in the fridge

Awesome man, thank you for the replies. I will use this method when I finally get around to agar. I don't grow a lot so my success with MS has gotten me plenty of fruit to go around. However agar looks fun, and I could potentially get a high quality consistent mycelium so I know what to expect from each grow, also there is the benefit of my flushes being more consistent which is really nice so I don't have to harvest for 3-4 days in a row.
I built a SAB from SpitfireJedi's tek, and I have tons of spore syringes.. its just a matter of time.

The number one thing that helps me is to use the antibiotic agar frim fungi, i use these grow bags http://out-grow.com/mushroom-grow-bags-c-13/small-mushroom-grow-bags-with-5-181m-filter-p-94.html I cup wheat to one cup water , pc for 2 hours at 15 psi and let cool NOW COMES MY TRICK ... I use a full plate af agar for each bag Its very important to sluce up agar very small and mix in the bag VERY VERY well to break up and losen grain and get every pice of wheat in contact at some point with the agar
Between the antibiotic agar and the explosion of growth from the mycelium no contaminants have time to grow as fast as the mycelium to do One week in the bags are fully colonized

Ok awesome. I will be using 1 QT mason jars (with organic rye grain), so I assume 1 full plate of agar for each jar? Also, when in the SAB, you shoot your spores through the tape correct? I assume you open air transfer the agar to the bags correct?

That's true as hell. I can't count all the old threads I've read where people where having good conversations or arguing or only getting a single response but they've all hit me in some way that was perfect when I needed it.

Bottles wear hard for me to mix up the grains and agar enough, I have a flow hood so everythings done in it Honestly organic is fine but i use Feed wheat 16.00 bucks for a 50 pound bag

Ok so I assume 1 container of agar per 1 QT of grain isn't stupid to do based on how much wheat you are putting in your bags. Also I will stick the needle through the tape when injecting spores into the agar container while in the SAB and not worry with putting a second piece of tape over the hole. I have also read that people just use a drop of spores from the syringe per container of agar (after heavily shaking the syringe of course).

Sounds good but id tape over any hole yiu make , better to be safe than sorry

why don't you try taking a drop of spore solution and put it on a wire loop, which you smear across the center in a line. You'll greatly increase the spore's chances of germinating without a contam right on top of it.
I wouln't leave an open hole in you agar lid, though. Put some micropore tape or something over it

Dont leave an open hole. Big no no!
And don't squirt spores onto agar, they will germinate all over as the water rolls around, making it hard to distinguish clean growth and/or may be grow into mold or bacteria colonies.
A sterile wire loop works fine but I prefer to use a loop to take spores from a print, when using liquid I think its better to drop it onto a sterile cotton swab (you can sterilize q tips by wrapping them into a foil pouch placed in a mason jar in PC cycle).
Swipe the spores on the plate in a zig zag- this makes it easy to identify clean growth.
If you are going to work with agar you *have* to learn sterile tek and how to open the lid. This is why I say not to bother injecting through the lid anyways since you are going to be making a transfer with a scalpel anyways

Ok so I will definitely not leave an open hole. I will be doing all transfers in the SAB, but the mycelium will already be alive and colonizing at that point so I assume wouldn't be as susceptible to contamination as would be spores trying to germinate in the agar.
I watched RR's video on transfers so I have a general idea of what to look for. I noticed how he emphasized not to let your hands or arms go over the top of or rest over the top of the lids as to lower the chance of bacteria getting inside the dish when doing a transfer.
I like the q-tip idea, I didn't even consider when dropping a few drops of spores there is just isolated drops of billions of spores so it would be difficult to separate contam from good healthy mycelium.
For long term storage of agar, what do you suggest?
