Making spore syringe from print Newbie Questions

Hi,
Now i am trying to make syringe from copelandia cyanescens print.
Is there any cooldown time after put those spores into syringe? I read that: The spores need time to hydrate in the water. This will take at least 24 hours, but 2-3 days is even better
Is it necessary?

IME just shake them up well and you're good to go. Never really seen a difference in hydrating vs using immediately.

Depends on how old. The older they are the more soaking is needed. A 24 hour soak period activates the spores in most cases. With really new spores, the chances of contamination are really good, and too old they loose their viability. I found that spores that are aged about 3 to 6 months are best as it gives time for the contamination in the spores to die, but most of the mushroom spores are still viable. Use a weak ozone source (enough to smell like fresh air) such as an air ionizer in a still air box and scrape them in the water, stopper and shake well. This will get most of the loose mold spores and bacterial contamination, but will not hurt most of the mushroom spores if done quickly. Hope this helps.

Honestly, if this is the first time working with sterility and a SAB, you should leave some spores on the print in case it fails. If it does, use that leftover print to inoculate some agar. You could just use the syringe but if you contaminate it during "syringing", it will be harder to clean up. Not to mention that spore solutions are generally stubborn about germinating.

The age of the print definitely means a lot like Eywa said. Never been one to take a print unless it's going to be used soon, that's the only reason why I never worry about rehydrating them.

I got a 4 year old allenii print wrapped in foil, put in a single Ziploc bag and stored in a dresser to germinate on agar with absolutely no problems that's probably the oldest print I've had.

Print swab to agar is the easiest and best option always, but a new person might not know that much. Either way if you go print to syringe, it still has to get cleaned up with agar or there's going to be a lot of failures. You don't want to make a syringe without cleaning up a wild print. At least that's what I think we are all trying to say in not so many words.

i feel that it makes no difference at all but it does raise the chance of contams developing further i prefer prints in most cases i scratch a bit off with a scalpel rub it on agar add one drop off water and done
