11 rounds of transfers shouldn't i be seeing sectoring by now?

So i started an escondido culture from spores on agar and these are the result of about 11 rounds of transfers and i feel like i'm not seeing defined sectoring like i should be by now. Any thoughts?

You probably should've used them like 9 transfers ago...but then again I'm not exactly sure what you're really looking for specifically...they look fine to me?

I also add gypsum to it but not necessary) Spitball's Monotub Tek (A liner isn't necessary but is useful) Use 6500k lights throughout the whole process. When you wake up, turn the light on. When you go to sleep turn the light off. It's as simple as that.

Sectoring appears on the first plate if you let it grow out a bit, I don't think I have ever not had sectoring by the second plate.

I'm curious to hear what others have to say about this as well.. I'm on my 5th round of transfers of one strain right now and they are all looking similar to yours. I just tried a slightly less nutritious mix of agar this round to try to promote more rhizo and hopefully sectoring, maybe you could benefit from trying that as well

I use a pretty low ratio, 6 grams of malt extract to 12 grams agar. I'm sure they would do ok if i just put em to grain but im really trying to get monocultures down.

If you took say wedges 1/16th of the outer growth on each plate and used all 16 wedges to make new plates each time you would end up with over 1 million plates by the 6th transfer. If you have done 11 transfers, you will have monocultures.

Also, it appears you are taking large transfers. You have some really beautiful growth! If you really want isolates, you must take small transfers--as small as you can. Like a dry grain of rice.
