Hi everyone, hope you are all doing well
I am about to do my 2nd round of transfers from multispore of a few different races
Wanted to get some feedback from Pasty and the other agar experts about which sectors to transfer, etc :
Plate 20-A2:

Plate 20-B1:

Plate 33-A1 (Accidentally dropped this plate after the transfer lol):

Plate 33-A2(Accidentally dropped this plate after the transfer too):

 Plate 33-B1:

 Plate 100-A1:
Plate PT (multispore pan trops, not transferred yet):

Sorry for the bad pics, they actually look a lot better in person
Just finished pouring 20 plates pf MYPA a la Stamets , planning to start the transfers soon. any input would be much appreciated

transfer anything from any of the plates that aren't a hot mess. toss the rest.

thats about what i figured, i already have a few spots in mind.
Knock on wood, my sterile tech must be pretty solid this time around, literally no visible contamination on the last 200-220 plates
the couple of plates i dropped look rough from the spread when it fell, but even they look healthy (more so that the pics make it seem)

i had 12 old spore syringes i found moving, and figured i would see if any of them are still good, so i made several plates of each, then started trying to isolate some strains of each of the ones that took off.
I plan to isolate 3-5 strains of each race and fruit each monoculture, keeping good notes on all the different characteristics a la stamets suggestions
i have really been enjoying getting back agar

Looks like you're off to a good start. Isolates are a load of work tho but goof experience that can really pay off. Good luck

thanks pasty you and RR inspired me, multiple times over the years haha
finished the second series of transfers early this am,hopefully some will be true monocultures
Next step is to make grain jars of each monoculture and make buckets or mini monotubs, and take good notes on the growth characteristics (been reading and re reading Stamets books and a lot of good threads to brush up too)
This time i made my own MYPA according to Stamets recipe and it seems sooooo much better than the premade powder MEA stuff i was ordering before. Looked a lot more like what i see in bacteriological and mycological texts hopefully it will perform well. My pouring technique was MUCH better this time too, with little to no condensation (which had been an issue before)
any advice in general for this battleplan?
