Timing of First Transfer, Multispore on Agar

hi everyone, been lurking for many years, i have learned so much from you guys, thanks a bunch to everyone here for all you contribute :smile:

So I have been germinating a few old-ish spore syringes i forgot about (2 yrs). About a week ago i made 6 plates of 4 different varieties of cubes, and I am starting to see some white fuzzy mycelium starting to develop where i streaked the spores :grin:

I am getting ready to do a few more plates, pan trop this time. The first plates are just now starting to show white growth, definitely not showing segments yet, only a small opaque growth beginning.

I think i poured the plates too hot last time, since almost all of them have lots of condensation on them. I stored each plate in its own ziplock and stacked them upside down, but the condensation was pretty immediate. Hasnt caused any problems yet that i can see, other than obstructing vision.

Should I go ahead and do a transfer on a few of the first plates, especially the ones with excess condensation? maybe try to get them onto a clearer plate so i can differentiate segments when it starts to get visible and less risk of contam? or should i wait for them to show segments before transferring?

I would wait to make sure the white growth is not a contam.

I took a couple of pics of one of the plates, everything looks really good to my novice eye, except for the condensation on the lid. These are 3 shots of the same plate:

 


also, here is a shot of the underside of the same plate. is the darkness ok? i think i used a big blob of spores on this plate that was visible as black even before the growth started, but it really sticks out against the white:



I have lots more with different levels of growth but very similar in appearance. should i go ahead and do a few transfers or wait until it starts to show segments?

I would let it grow out a bit or you risk transferring too many strains which take more plates to get down to isolates.

First plate - germinate spores
Second plate - transfer desirable sectors
Third plate - confirms if you have isolated strains and if so transfer to grain and slants.
