I use plain surgical tape and have basically never changed them. The paper towel provides a good cover for it and they don't get fucked up. I've never had satellites or any bacteria in my fresh plates and I usually store the bulk of them for months and months before using them.

This was probably answered before but I gave up browsing by page 35 so I will go ahead and ask: can I use just any type of honey? What I have on hand is that grainy, opaque honey I use for breakfast, can I use that or do I need refined honey? thanks

I'm sure that your honey would work just fine, its the simple sugars in it that support the mycleium's growth. However the best honey you could use is local honey from a nearby bee farm or something. Following that, you just want a lighter colored and less refined honey. Less refined=more simple sugars

great tek glad mini rounds were my first actual success with agar and i have been fascinated ever since 
Ive been reading the agar threads from people more experienced trying to figure out something pretty basic that i somehow missed:
When germinating multispore on agar for a strain isolation, do you wait for clear segments to show up before doing the first transfer? or is it a good idea to transfer to a fresh plate with less condensation on the lid?

Transfer from the spores as soon as you get clean growth, when the colony is about the size of a nickle. You won't see sectors until you do several transfers anyway.

You transfer it a few times if you wanna do isolation. Some don't even begin to isolate after a bunch of transfers so the sectors are more defined. Personally, I started to seperate them once I could make out the sectors but I see now that this uses up agar needlessly. Instead of having a couple of plates I have 10 plates with 4 cultures each, always needing to be transferred.
If you leave them until the sectors are easily defined, you will be saving a lot of agar in the process, not to mention time.

First of all, do not wipe after flaming. Alcohol sanitizes, flame sterilizes.
Second, you lightly depress the syringe to allow a little solution to cool the needle and a drop to fall onto the bottom of your SAB. The next drop goes onto your dish. You can also use the hot syringe to melt a divot into the agar to drop your spores into as well.

Should skip the wipe altogether. I never wipe my tools with iso unless I'm about to begin a session. Flame sterilization is all you will ever need.

 The first pic is on its 5th transfer. The second is on its third. I had taken transfered from the second plate a couple days agao.

Yeah, those look healthy enough to use as inoculant. Just let them grow out almost completely and inoculate some grains with wedges.

Cool, I've been using the agar recipe from munchauzen's 123 recipe so I could try out the blenderless liquid inoculate tek.

It rocks, dude. Munch is the man! That tek is so freaking easy and quick. Gonna be able to see the results soon enough. I'll be home from the beach to check it out in a couple days =]

Personally I would take a few small wedges from 10oclock to 12oclock and make plates from those, then use those to innoc. edit - from the second pic
