Is this myc growth?
    #23415320 - 07/05/16 10:18 PM (7 years, 7 months ago)

Hi, I have started the cultivation process by placing tissue samples onto nutrient agar. There are three pictures I wish to have someone review. The one with alot of dark brown is tissue from the gills of p subaeruginosa, I see a little white blob off the the side of the brown.

The other two, one is mycelium taken from woodchips in which p subaeruginosa was found, I scraped a few pieces of tissue off into the agar, this is why you see a few specs of wood in the myc.

The last one is from the stem of p subaeruginosa, it seems to be growing at a slower rate than the myc tissue sample and has a slight blue tinge around where the stems were placed.

I just want confirmation that this is myc and not bacteria, thanks.

transfer it to a new plate and you should get a clean culture.

:whathesaid: That's the only plate that has anything half usable.

Cheers dude, i'll cut out an agar wedge with that myc growth that you circled and transfer that to another plate. I will do the same when the stem plate gets to this stage as i'm not 100% sure that the mycelium in the plate that you circled is subs, however the myc sample was taken from directly beneath the sub patch. I'm going to discard the decayed gill plates as there is almost no growth.

Thanks once again!

Sorry for the second reply, but do you have any tips on transferring it to another plate other than making sure everything's 100% sterile? i.e. should i try and cut the cleanest looking part of the mycelium out

It's possible it's not subs, though I'd say it looks very promising. how fast did that myc grow? It's often a dead give away if it grows way too fast.

Just take a wedge from the outer part of the best looking growth. The bit i circled in red is what I would aim to cut out and transfer.

I placed the myc on the petri about 3-4 days ago. The stems were placed on a petri dish approx 1 day after

just make sure you flame sterilize that exacto before cutting in! dont want to instantly contam the one piece of good myc youve got :p

personally, id probably make 3 transfers off that one nice circle of myc. pick 3 different spots and see what happens.

good luck
