title. i feel like a little kid waiting for a Christmas present lol. i want so badly for my first jar to finish so i can start working on agar and do my first G2A. SO CLOSE!! (maybe a day or two)
i also have about 28 jars due for a shake lol. i let them go a little longer than planned, so today will be the official day for shaking ALL of them. right now EVERY single one of my jars has perfect clean mycelial growth, so im happy as can be. no signs of bacteria so far, fingers crossed.
ahh i cant wait to start isolating strains and transferring a bunch of agar lol. i have 5 strains total. gonna isolate strains of each ideally. and have multiple mono-tubs of each as well >;) (and of course i want to keep the cycle going this time by constantly transferring to new grains/bags of grains/agar/LC/LI/tubs/GH/other experiments!)
and then once a majority of jars are done, time to make some FAT 66qt mono-tubs haha. do you think 1:2/2.5 ratio with 6Q spawn and 12-15Q substrate will be enough for these tubs?
i usually work with smaller tubs, but im going a little bigger this time around since i have the spawn to work with. pasteurizing all that substrate is gonna SUCK though..
anywho, i went off track, but any answers would be helpful lol. im basically just trying to say...
TLDR: I CANT WAIT TO START DOING AGAR AND CYCLING TUBS!!! YEAHHHH BUDDY!!!

For my 66qt monos I use 6qts spawn, 1brick Eco earth coir, 2.5 qua vermiculite, 5.5qts water and a large handful of gypsum. A Ways get great colonization times and yields of course....
And just to be a stickler....you have 5 varieties you will work with to isolate strains from....B+, Golden Teacher, AA+....etc...these are varieties not strains....like if mushrooms were people....American is a variety and every individual American is a strain.....
I recommend cloning from a fruit body before you isolate, that gives you a much better chance of isolating genetics that will produce....if you just start with mycelium and randomly pick a strain to isolate you might be isolating bunk genetics....the good news is that the spores available to us commercially are so thoroughly artificially selected....bunk genetics are rare.

Yeah I use 5 quarts of spawn, 650g of coir, 3 quarts of verm, 5 quarts of water, and 1 cup of powered gypsum for the 66qt tub. This is what PastyWhyte does as well (minus the gypsum, not sure if he uses this or not).
Also, its very easy to throw 3 quarts of verm, a brick of coir, 1 cup of powered gypsum, and pour 5 quarts of water at 190 degrees in a bucket and seal the lid - not really a pain in the ass at all 
P.S. I take the water off of the stove at 190 degrees, so once the water hits the dry ingredients it cools off to probably around 165-170 pretty quickly.

waiting on agar materials :p but i might do just that as soon as its here. hopefully tomorrow. plus i think i have a jar that just hit 100%. colonized just a bit quicker than the rest. but ill be spawning to bulk soon!

ah,okay, just this that threw me off thinking you had your sterilized plates all ready watiting for the bottom of a grain jar to complete

i write things weird when im stoned -__- which has been too often since i started making my own dabs and waxes haha(super fun by the way).
i went ahead and purchased 12 borosilicate glass petri dishes. i want to try a pour tek, and the no pour tek nearly simultaneously. sort of a test of whether or not my sterile technique within my SAB is truly as good as i think it is :p
still gotta grab me some pp5's for the no pour. but im gonna use both MS's i have for 2 plates, and then whatever grain jars finish first will be first to transfer onto plates as well. those jars will then immediately go into bigger grain bags or straight into a bulk substrate if enough jars are ready (;
that should start me on a fairly good cycle of tubs right off the bat. ill have enough myc growing consistently to keep the tubs flowing id imagine at least i hope. this is all under the premise that nothing goes horribly wrong hehe.

yoo who bought all the small pp5's in my area? haha went to 3 local stores and found a total of 1 pack of mini rounds by glad, and a set of rubbermaids(they are square but i doubt that matters much?). wth guys.

Personally I prefer the 4oz Ball storage jars with the plastic lids....durable....clear....flattish bottoms....:
 
Love the plastic lids....only used gladware once....didn't like it

Do the colors mean anything or are they just rainbow for fun? I used to color code mine for different mixes and whatnot.

yeaaah buddy, i like what i see haha. all good info though guys, thank you.
my agar supplies are officially here so im excited(i have left over GLME from a previous attempt at agar. super old but i think its fine. i had it vacuum sealed lol). and then the agar of course. starting 2g LME + 2g Agar + 100ml water; see what that gets me in terms of dishes and consistency and go from there.
my jars colonized well too. might update my grow now with colonization pics or wait till i start work on agar. but either way im getting it going tonight/tomorrow depending on time constraints!
i will have PE + GT going MS to Agar; B+ going Grain to Agar.

just telling the ninja turtles with the color in that pic....orange, red, blue, and purple
Now a days I like to do red because I lost my blue color and can't make purple anymore....but i like to make the plates dark red or purple because my eyes aren't the best and it makes it easier to see mycelium on the agar for me

just an opposite opinion here...
I like the plastic mini-rounds because I can actually squeeze the plate to release the agar and drop it into grains or LI water, inoculation with no tools or flames. I've been using the same ones for almost 2 years now, so I would also consider them durable. I've always been able to spot contams in them, and if I want a closer inspection, I just take the lid off in a SAB. Not sure what the flatness of the bottoms has to do with the price of rice in china. The working surface of the agar is always flat regardless of the vessel.

The flatness of the bottom has to do with pouring the plates....for instance in the half pint jars the bottoms are very convex so when you pour it the plates, they look thick enough from the side, but the agar might not even be covering the middle....so by the time you have a good depth I. The middle, the outer edges are extremely thick making the wedges harder to handle...IME.

I also prefer plastic (lol) for not only the reasons munch stated (tiger dropping the plates is super cool, hell someone just did an agar sandwich that way the other day) but also because glass half pints are taller and I hate feeling like I'm reaching down that much. I also find glass slippery to work with in the SAB.
At the end of the day it's all about preference. I have always encouraged people to try everything at least once and decide for themselves what they prefer.
