When should I use the syringe?

Hi all!
I'm a new member here, but I did read most of the rules and did search before posting, but didn't find a specific answer to this question.
The thing is, I used my spore print (cubensis) to make a syringe (sterilized everything, used distilled water, etc.). It's been about 12 hours now, and I'm not sure what should I be looking for? How do I know if my syringe is ready to use in a jar, and are the spores alive or hydrated?

Yep it's ready. It doesn't take long for spores to hydrate. Some say to let it sit for a couple of hours, but it's ready immediately imo.

Alright, I can finally update my thread!
You guys were correct, the syringe I made worked nicely, and my jars got inoculated, and since a few days ago they have been fully colonized! All 3/3 are strong and healthy, thanks to the members and the vast amount of info on this forum 
Now though, I have a few dilemmas.
I bought perlite and a large plastic container as suggested in the Teks, but I'm not sure how to customize my fruiting chamber.
- (most important) Do I need to drill holes on all 6 sides, or can I just keep the top opened and covered with a large plastic bag which I can vent 1-2 times per day? Wouldn't the holes eventually contaminate the perlite and the cakes?
- Do I drain the perlite before putting it in the chamber? Does this imply I need to mix the perlite with water in a different pot and then transfer to the chamber?
- Should I dunk my jars, or since this is their first flash, I don't need to? Also, does dunking require boiled/sterilized water or can I just use distilled water in a jar?

Open air does not contaminate a grow during fruiting, put holes on all 6 sides. Put perlite in tub, dump water in tub, drain water out of tub. Dunk the cakes.

That's a nice guide, I'll be making the FC according to that tek.
One final question though: Some of my cakes seem to be growing through the verm layer that was put on top, and it seems washing the cake with water and a towel doesn't remove the whole verm layer. Should I use a knife to cut the top off, or would this open up the cake to possible contams?

Just rinse off what you can rinse off with water and your fingers, and leave the rest.
