i try to do it within 24 hours.  even though you sterilize your grain, it won't kill all the endospores and you may end up with bacteria if you wait too long.  you want it to colonize as fast as possible :thumbup:
also, you should put those spores on agar first, then drop a clean wedge of mycelium into your grain jars.  you'll have much better success starting with a clean culture.  spores can be really dirty. :thumbup:
