none of it looks like oyster mycelium. oysters have much thicker mycelium. those grains barely look colonized. they are probably colonized by some kind of mold.
That's what I was thinking but it's so strange. I'm tempted to let it grow out and see what happens.
Don't know what happened on this last run. Maybe the PC didn't stay at a consistent 15 or maybe with the addition of bags in the run weren't getting enough steam exposure and therefore could have used longer PC times. Lot of endospores in my amateur opinion (the growth didn't start at the top or at inoculation point but just kind of all over).

Mold is not from insufficient sterilization, 130f sustained for 30 min will kill most molds. I would be looking at your inoculate myself.

Hmmmm, haven't had this much trouble in the past and this was the first time I used my buddy's flowhood- that's the only difference. Their alcohol flame was smaller than mine, and perhaps I moved too quickly in my newly found freedom. All of these were wedge drops and the agar culture looked clean as a whistle. If it's not endospores it had to be one of the three newly introduced variables. Perhaps it wasn't achieving laminar flow? Though I'd think they would have figured that out by now. I'd like to narrow it down though so I know exactly what I'm doing wrong. Every contamination is an opportunity to learn.

I had a quart grow trich, near the bottom of the jar after sterilizing at about 17 psi for 2 hours, this was like 2 months ago. My innoculant went bad so I had nothing to inoculate this batch of jars with, so they just sat around for weeks unshaken/undisturbed. Ok it doesn't happen that often but I've seen it happen. I can't personally rule out mold just because temps were over 130F... based on more than one experience.

Filter fails and bad procedure then. Get your PC gauge recalibrated lol. But mold is killed by pasteurization temps. We sterilize because of bacteria and pretty much only because of bacteria. I have seen uninoculated jars get bacterial over time but the only time I ever saw an unopened and uninoculated jar grow mold was a filter fail.

I'm guessing it was my technique in front of the flowhood for the first time. I was so excited the old school bartender came out of me and I flowed through a ton of work really quickly, may have churned up too much air and disrupted laminar flow. It was just so exciting to not be in an SAB, lol.
Quick question Pasty, I'm about to supplement for the first time and I was wondering if % was calculated by volume or weight to sawdust?

I would lie and say I do by volume but even that's a stretch. I tend to use grain as my inoculate for most shit so I don't suppliment stuff much, usually I just up the spawn ratio.
I'm pretty ham fisted with my bulk subs and such, if you were looking for something specific to a certain shiitake strain or something, I recommend the guys over in G&MM, they got their shit dialed in tighter than me.

Still rocking the GT.
First set knocked up 5/10 and birthed 6/6. The new jars that just got dunked and rolled were knocked up 5/20 and birthed today.
I really am digging the dunk, rinse, roll and neglect for a day. Most if not all the verm stayed on the 12. Rehydrated perlite once again so I added an inch and a half, fixed the draft from the door and moved a room fan. Enough of this shit lol.
Ideally by the time the first set is done fruiting, the second set should just about be starting. I'm seeing small hyphal knots and spotted 3 pins. Then it's bye bye SGFC, woot!

Don't toss the SGFC tho. They come in handy on occasion. But monos are where it's at for sure

Yeah they all seem to be in bed though at this point, lol, and this is some of the only spare time I have so I'm getting it done- I'll do volume- a few of the big wigs over there use 18% so that's what I'm going with. I have nothing to compare it to- but this is also the start of my copious note taking through a spreadsheet that I hope to make accessible online so others can see for themselves and maybe even add their own sheets.

18% volume seems about right for most species tho I would error on the side of less with shiitake. Oysters on the other hand just don't give a fuck, they can fruit off cube subs lol.

I figured to just make a smaller one. This is great and all and can fit 30 comfortably, but is quite the eyesore. I'll need a smaller one once I get tissue to agar, run a few tubs, LI some for performance and later down the line test potency off the cakes. I still have some reading to do on that but I believe it's the direction that people head when isolating for both potency and speed.
I've got a lot of culture work this weekend, then it's in the 100s for a while. The first round of cakes is approaching the point in which misting won't be very necessary anymore. It worked great last time.. but you saw my first run with mutants, cracked caps and bacteria from when I was drowning them.
Loving the upside down jar lids too. No pooling water, no foil, better micro climates.

great info on the cloning, Pasty. I have always taken from the lower, and a few recent ones have not done so well off the bat. Gonna try from near the top next time. My problem, i think, is that i waited too long to take the sample, anyways. they were very mature, just because i had no time earlier. So getting a more vigorous sample would help. Thanks!
