Thanks Pasty!
Good simple agar tek, made some a few nights ago, easy to read and easy to follow. I had to use different style containers but otherwise followed the procedure to the letter, good stuff.

I see some clean growth that has separated from the bacteria. get in there and transfer that ASAP, otherwise you might have to do an agar sandwhich to save that

I'd say around 8 o'clock on the last photo. Take the smallest piece of mycelium posible. Like a quarter the size if a grain of rice.

4th picture. 4 oclock - 11 oclock.
agar sandwhich is to just take a 2nd agar plate, and drop the agar puck on top of the first one. the bacteria can't grow up through it, but cube myc can.

Bloby said: I've just bought some of these (link) pp5 containers so I can have a go at this tek. I have just one question, in the description of the containers it says "Always leave the lid 1/4 open when sterilising." I haven't seen this mentioned anywhere else so I think ill try just four plates 2 open 2 closed and see what happens the first time around.
Does anyone else have any info on this?
Is that company reputable?

How long does it usually take the myc to grow through?

Do you think it'd be easier to transfer away from a different starter plate?

I have 8 plates I used a swab on. They haven't shown any contam or growth.
The myc above is from a jar a spawned, it was leftover in the bottom and had begun to pin so I tried to transfer it to agar to save it. If I can't I'm back to square one. I have some syringes but I never have any luck on agar with spore solution, my plates for whatever reason always have pooling water. Even the ones above sat a week before transferring anything to them to try to fight the condensation/pooling water. These are all on Lightmalt/water/agar

I use potatoes flakes/agar/water. I also started with live myc from a pf jar because my initial try with spores never took off
My biggest issue from my first plates was pooling water also. I started bringing my pressure cooker up a lot slower...a loot slower, and it seemed to help quite a bit with condensation.
I've had had some issues with bacteria on my agar as well. I usually just open it up inside my sab and cut the bacteria out of the agar. Probably not the best advice but it's worked for me.
The way I see it, you have myc growing on agar. As long as your plates are sterile and you don't leave the lid open for a million years to transfer then your far from square one.
If there is even a small sliver of myc you can transfer away from that contaminate then you should be good to go.
It may take a few more transfers but if your persistent you'll see mushies in your future.

That was my first time using them but my containers arrived in a couple of days, well packaged etc. After doing my test run of 6 plates I brought the pressure up super slowly and allowed to cool before opening the cooker and non on them have warped (even the plate I left closed). My cooker only goes to 11.6 PSI so this might have had an influence.
I'll report back once I've used them a few more times, also its been 4 days since having a play in the SAB and non have contaminated yet so fingers crossed the seal holds.

A bunch of my PP's got fucked with bacteria when my fridge's cycle went through the increasing summer heat. I probably should've be storing plates that long anyway.
Anyway, I don't have lunchboxes lying around for insulation. What do you guys think about stacking them in glass mason jars?

What? I'm talking about protecting the myc from fridge cycles, not making transfers.

I don't understand what's happening... temperatures shouldn't add bacteria just make it easier to grow... leaving it in the fridge shouldn't change a thing

The temperature changes cause water to start pooling which can't be good for anything. In any case, I need to shield them from the temperature changes which is what I'm asking about. Ziploc bags don't work.
