I cooked some plates up on Tuesday and have them stored in a small tote, are they still good to go?

If they didn't even last 4 days they would be pretty useless for this hobby.  So... hell yeah they're good to go.  Would probably be the same if you asked again in a month too.

I somtimes leave plates in a tote for months before inoculating them. As long as they don't dry out, they're good to go. Generally I let my plates sit for at least a week to let the condensation clear up before I inoculate them.

When transferring to agar from qtip should there be visible spores on the agar after I swipe?

Not necessarily. The smallest speck visible is around 10 million spores.

I've just bought some of these (link) pp5 containers so I can have a go at this tek. I have just one question, in the description of the containers it says "Always leave the lid 1/4 open when sterilising." I haven't seen this mentioned anywhere else so I think ill try just four plates 2 open 2 closed and see what happens the first time around.

Does anyone else have any info on this?

i dont leave my lids open at all. i just assumed the hole was enough for it not to implode on itself =p

seems to be working fine. have done a few of the glad minis about 4 times now and not one has started warping or anything.

i dont recall reading to leave them cracked haha.. but i probably either missed it somehow or forgot.

I've worked with those, I don't like them at all but you can use them just fine. The filter patch won't do anything to prevent  the containers from crushing during the sterilization run, so you have to drill a hole (or melt a hole) as per the tek.

Looks like I jumped the gun buying these. What about them didn't you like? I'll just leave one of them sealed to test, my cooker only goes to 11.6 PSI so maybe I'll get away with it. I wonder why these are happy to crush whilst jars are fine?

The filter is just finer, and can't exchange gas fast enough to prevent crushing or popping the lid off.  I've had Whatman filters do it to mason jars.

Those would be great for bottle substrates.  Just loosen the lid,  cover with foil,  and tighten with the foil on once you open the PC.

They are very big for this, the plastic is very very thin and will warp at the first opportunity, and the hermetic seal is a bitch to open in the SAB. Also, the filter patch is useless, since you're going to drill a hole anyways and if you don't drill holes and leave them open during the sterilization run, I advise you against leaving a crack open because they'll warp all the same, instead pop off the lid and leave it loosely on top of the container (see how sketchy this idea is?)

I can give you the result of the test already. It'll warp and you'll have to trash it. Even if you were to just steam-sterilize them without any pressure, they'll warp all the same.

I transferred some older rye from a jar that had pins on it. I had some trouble with droplets of water on agar.
Just trying to sort out of there's contam growing on the plate now as well.
I plan to transfer again today just incase.
