Light helps but isn't necessary.

I'd personally kick that temp up between 72-78°f for faster growth.

since you ran so many, how thin can i go on the agar pours using the 4oz ball jars without ill results/ normally run about 1/4" - 1/2"

can i run 1/8" w/ same results?

We both know ima try, wanted your thoughts

BD, I've found if I go too thin, that I have found that it's often much harder to use as a receiving plate because It can be harder to get your sample off the blade.

idk about deth but i use 125  water 5 grams of mea to make 10 4 0z jelly jars the deth i have never mesured

thats about how deep they are

I'm trying to figure out what I did wrong with a batch of plates. All my cultures are growing ridiculously slowly. The picture is just one example.

I took this pic today. It's a transfer I made from a clone grown invitro. This transfer was done in April 15th. I ruled out senescence, myc expanded very little before I took this sample. I also ruled out the possibility that all my cultures are this slow, I mean what are the chaces of that happening? (sorry for the shitty quality)


-I did this batch using two layers of micropore tape, but I don't think this is the issue here at all.
-Normal agar mix (20 g --- 1 litre of water).
- 1/2 teaspoon of honey.

I think honey is the culprit. I never weigh out the honey and I think I just put too much. To be sure if I put too much honey, today I weighed out a teaspoon and it is ridiculously heavy!
RR said that honey is antifungal, so can this be the issue?
Can I use raw sugar instead of honey?

That does look over nutritious. Just a drop of karo is what I say but you could use SITR or honey too. Just don't use too much. If SITR I would keep it at under 1%.

Honeys antifungal properties will only be for natural honey not the colored sugar water you buy in the store. But at the concentrations we are looking for its hardly an issue. Follow the tek, just a drop is all you need.

wait, so you are making straight honey agar?  No potato, nothing but agar and honey?  If so, that is probably your problem.

Try pasty's tried and true PDA that is on first page of this thread.  Worked for thousands of plates for me. 

I just switched to MEA at 1 tsp ME, 1tsp agar per 1/2cup water. Just made first transfers to those plates yesterday so too early to tell.  But I felt I needed to give my cultures some new feed...

Thanks Pasty. Oh fuck! I'm using natural honey, as natural as it gets. I'd switch to karo if only I could find it, it's going to be SITR then.

Okay wanted to ask a pretty noob question so bare with me. I want to use pastyplate agar tek. So i have agar agar bars just like shown in the pic , Idaho mashed potato flakes forgot to get some karo so i'll be just using honey(I hope any honey would work the one i have has no brand local made my man) also bought 10 glass petri dishes which few of them i opened from time to time to show my mom lol . So question are , i'm using glass petri's so i can't punch a hole in them for obvious reason so how do i micro tape it and sterilize the agar agar once i've made agar and do i need to sterilize them before as i've opened and touched them or can i just soak them in bleach solution pour agar agar and then sterilize. This is my first time growing and i'm starting of with agar agar for the best results. :smile:

Real local honey has antifungal properties.. You need fake sugar honey.  You can't use glass petris with this tek.  You cannot pre-pour glass petris at all.

You pour petris in the SAB or in front of a flow hood after sterilization in a larger vessel... Look up a pour tek if you're set on using glass petris.  And no, bleach has no use in this hobby aside from normal cleaning in between grows.  Only heat can sterilize.

You can use sugar.  Somewhat complex sugars are better though.  Any honey that comes in a bear shaped bottle is almost always not 100% real honey.  And yes you have to pressure cook the glass petris and agar separately, then pour the hot agar into them in sterile conditions.

at trader joe's they sell this shit they label honey, but one taste and you can immediately tell its molasses and corn syrup



this is about as fake as honey can get. oh ya the FDA said this was from China, not Turkey, either. idk how they still sell it.

Because it's like, a product that's allegedly supposed to mimic the taste of actual turkish honey.  Like uh.. you know, swedish fish.  We know they're not fish and they aren't from sweden.  Except in this case it's pretty deceptive.  But that's probably the kinda bullshit they're hiding behind for it.

In the pour tek he is using agar agar flakes and light malt extract neither of which i have. I've already gone through alot of hassel the supplies I have as many of them aren't available locally. I have agar agar bars mashed potato flakes and house hold sugar if not honey. I have glass petri's and a pc i also have a steel stand to put inside my PC to allow space between water and glass petri's. I also bought heavy duty Foil. Is there anyway to work , I want to use this method with my glass petri's .

Synthetic honey aren't easy to find here if at all.  I'd be better off buying a corn syrup (would any brand work of any formula?) Can i use plain simple house hold sugar instead of honey or corn syrup?
