transfer questions

All the pastyplates I made up now have tomentose mycelium growing in them.... my first question is: is this normal from spore inoculated agar? My 2nd question is: how do I identify sectors in tomentose myc? My final question is: how do I promote rhizomorphic mycelium on agar so that I can identify sectors?

It could appear tomentose simply because of so many genetics, it could also be bacteria, or even having a very nutritious agar.
So, first question did you use a lot of spores? If so, just just make transfers to see what you get. It may take a few.

I believe it's genetics, the cake that the fruit I printed was on had tomentose myc too

No just a very tiny bit, the clump of spores I scraped off was about the size of a flea

I used the agar recipe from Pastywhyte...with a tad bit more agar powder for firmer agar

Yep. It's a process. It's hard because you can't see the spores with the naked eye. But it just takes some practice.

You normally start to see sectoring after quite a few transfers from MS, it can take 5 if you're lucky or even more than 10.
Also, don't worry about tomentose myc. Myc will become rhizo if the conditions change.
Just transfer some samples from the leading edge of that MS plate to new ones, taking that the myc looks healthy.

It does look nice and white....I'll keep taking transfers and using the agar sandwhich tek so I can clean them up if need be....

Can you wait too long to transfer?
I have 15 pastyplates that are about 3 weeks old now. Can I use a 3" blunt tip 14 gauge needle as my scalpel?
Flame sterilized ofc. It's all I have right now. You only need like a mm piece to transfer correct?

I was told it was best to wait until the myc was tge size of a quarter, I don't know if that helps you out any bro?

heh, i let a few of mine wait a little too long as well. making the agar tonight and transferring a ton in the morning. =]
was going to do it a few days ago, but i got sick blehh

No need to sandwich it. The sandwich tek is normally used to clone wild fruits and when you're dealing with heavy bacterial contamination or even if you need to germinate some stubborn spores.
