Jar Design
    #23189332 - 05/04/16 04:51 PM (8 years, 8 months ago)

Hello all,

So my first batch I think I messed up. I just used foil as a filter with 2 holes slightly bigger than the needle of the spore syringe for a quart jar with rye seed as the grain. I inoculated them 3 days ago and do not see any progress in terms of mycelium or contams. I know the key to this patients, so it is not like I'm expecting results by now but I would like to improve the methods and environment I'm cultivating in. (I based my grow technique on the TMC and some older techniques I found on here)

I did some searching in regards to jar filters and found a lot of results and most where 3+ years old and to avoid of doing the wrong older method again. I wanted to ask the pros for some feedback.

I would like to to go with the polyfill technique since it is easiest to come across, I was just wondering if that is the best choice for grains? Since I will be shaking them in order to loosen the clumps up. Or do you not shake until the end?

So if some of you guys could post your jar design in terms of filters and hole size that would be fantastic. Since I will be redoing my cultivation based of these results and with some agar work.

Thanks again

- Mike

Two layers of micro pore tape over a ge hole is easy and effective.
The foil doesn't sound effective at all.
Pastywhyte has been using no filter, just slightly loosened lids with success; I have started doing the same. You just have to remember to tighten the lid before moving or shaking the jar. I haven't had much luck with poly in my lids.

:whathesaid:

I'm gonna have to try this loose lid tek. I only use polyfil lids when using G2G as I make sure to have a nice layer of mycelium covering the top. Acts as a barrier as it starts to colonize and have had a lot of success. I've had many failures when trying this with agar to grain.

I can't say too much because this is my first grow also. Originally I was going to have lid with both a SFD and a SHIP, inject my spores through the SHIP, and then rock and roll but everybody convinced me that spores don't do so well when they go straight to grain.

Instead I bought plastic lids, placed a 1/4 inch hole right in the center, cut out a quarter sized circle out of a SFD, and then applied it to the top of the plastic kid for my 1 qt jars. In doing so, I won't be able to inoculate with spore syringe, but it forces me to use agar.

Now I put my spore syringe to agar that is prepared inside of a glad mini round via "pasty plate"(some people swear against spore to agar) then once my agar is fully colonized, I use the "tiger drop" method to drop my agar into my 1 qt jars that have the SFD for GE. The idea behind this set up for me is to only order a spore syringe once. After the agar has gone successfully to the grain, I will do G2G transfers, grain to agar transfers, along with cloning fruits to the agar.

So far it is working out very good. I brought 18 agar dishes to my grains in about 20-30 minutes, with no signs of contamination yet. I think this will be a very successful first grow.

I probably drove people nuts with all the questions in the mean time though haha

Foil is not a filter. Foil with a hole in it with no filter material is going to fail. If you own a SAB you do not need a SHIP. Simply crack the lid and drop in wedge, colonized grain, or squirt in LC or spore solution. But foil by itself is not a filter, at best it's a barrier.

Yes I recently just completed another experiment with plastic lids cracked open a touch. It's pretty damn easy.

Sorry for the confusion. I meant to say 2 holes in the jar lid not the foil itself. What is SFD? So spore microtape should yield good result then. Have any of you tried the post office Tyvek filter method? Thank you for the great posts guys!

I will be going with the pasty plate method as well. I will be prepping it today and trying it out tomorrow. What is expected time of colonization?

SFD=synthetic filter disk. I got about dime sized growth on my pasty plate after maybe 3-4 days? I am horrible at keeping a log so far. Then I cut those dime sized growths into quarters and transferred to more pasty plates...they fully colonized in about 7-10 days, and then they were dropped into my grain jars that have the SFD on the plastic lid...but apparently you don't need SFDs on the plastic lid and can just leave the lid loose. Oh well, mine are already made so I'm going to stick with them lol

Using plastic or metal lids?

Could you also use this with the metal lids? I want to try but seems like there would be too much of a seal to allow any crack for GE. Thanks Pasty.

I used plastic. People used metal also. In any event, you need to lightly sand where you're going to attach the SFD with silicon. The problem with the metal lids, is that sanded area rusts more easily. I haven't worked with metal lids, but I like the plastic ones because I can take the lid off in one motion. The metal lids have the 2 part lids...if that makes sense?

I don't think the loosely put on lids would apply to metal lids, but I'm sure pasty will give you a better answer.

Makes sense! I'll just go ahead with the micro spore for my first run and explore options from there
