Hey all! I'm trying to contain my excitement here and not get my hopes up too high, but I think I actually got my spores to take finally and germinate! ...but as it's my first time doing agar (or cultivating at all) I wanna just confirm it. I'm doubly suspicious that it's working for other reasons as well; once I explain my situation you'll see why. But first here are the pics. If you care to read the story you can read on after the ID part of this post! That way others don't have to if they just wanna ID real quick.

I know there are lots of contams visible on the agar, these appeared much much earlier after inoculation. From a wild print and possibly unsterile syringe. 




(Forgive me if one of the pics is blurry, I'm doing this from my phone and it's hard to see which ones I uploaded)

The fact that where it's centered is off white makes me worried, but dammit it looks too much lIke myc to my untrained eye to not get my hopes up a little after I'd almost given up.



Okay now for backstory if anyone cares.


When I got interested in trying my hand at cultivation many months ago, my heart kind of got set on the idea of cultivating from a local source of cubes. Well, actually, even more specific than that. About 3-4 years prior, I discovered and then used shroomery to identify what looked to be cubes growing in my backyard/pasture (I have a horse... probably makes most of you jealous!) And indeed they were, and I spent the next 2 weeks collecting them. Even though I let them mature and disperse spores however, they never came back again following years.

Fast forward to now and I still have a lot of spores, stored out of light and in okay temp. But not at all sterile nor stored neatly or cleanly. Despite that and despite how difficult cleaning and isolating them is, I've researched and am giving it a go.

Weeks prior i had already made a syringe of spores since they were so old, and needed rehydrating. I made agar and my first batch was successful; uninoculated plates have developed no contams. But both times I inoculated plates, they quickly developed contams, mainly bacterial I believe- like that pink gunk. I read and found it takes 3-7 days for spores to show, and hoped the plates wouldn't be taken over prior to that happening-if it would happen.

It's been about 6 days since I inoculated and honestly I'd pretty much given up. I thought the spores might be too old or stored improperly and were no longer viable-- to the point that I actually ordered a syringe of penis envy 6 last night, something I was trying to avoid. Then today I decide to look at it again and what do you know, new growth! The contams had stalled and the plates had been pretty much stagnant the last several days so I hadn't been checking as often. I won't be surprised if someone tells me that it's just a bunch of mold thats gonna sporulate in 24 hrs, but dammit I'm hopeful and if it is myc I will be so proud. Thanks for reading!

Seems like mold to me. Didn't read all your post so forgive me if you already said this but, is this from a vendor? Or is it a homemade print/syringe?

Edit:

you should take pics of mycelium germination with a light on the other side of the plate so we can see better what is going on. That said, you can probably find what germinating spores look like if you use the search function.

If you can find them again you'd be better off cloneing tissue.

Yeah, it was just a dream idea to do it from those old spores since nature put them in my own backyard, lol.

Glad I went ahead and ordered some actually clean ones, those will be a piece of cake comparatively I'm sure.


I think the edges of my dishes are poorly wrapped. I don't like using plastic wrap. I know everybody says parafilm, but what about micropore tape? Anyone use that?

I have only ever used parafilm, it's annoying when it tears and you have to double or triple wrap but it's easy to use and a roll lasts a long time. If I didn't have any other choice I would probably use plastic wrap until I get a new roll of parafilm but micropore tape would be the last thing I would wanna use. In reality, petri dishes don't even need GE to colonize a whole plate so you can use anything that seals well but parafilm is too comfortable to forget about.

You can try cutting rolls of plastic wrap to make them small enough so you don't have to play around with the scissors for a long time. Some people swear by rolls of wrap cut in 4 but again, I never used it and would like not to.
