Too wet? BRF Cakes
    #23182477 - 05/02/16 11:51 PM (8 years, 8 months ago)

I made some BRF cakes and the mixture seemed fine but after the pressure cook it seems that water got into the foil and made many of the jars soaked. I said f it and inoculated them anyway. None have any contaminates because I did it very sterile and used 20cc for 12 jars. Some of these look too wet. Let me know what you guys thinks and what I should do? I have 12 more jars sitting next to me in my PC waiting to be used with my last 10cc syringe. (I will only use 10 jars).

Also this is about at day 10.

They're looking pretty good, if you're worried you can leave the jars slightly loosened to allow for some better air flow. Moisture buildup can cause contamination but once your cakes mature a little longer they're better at fighting off contams. How long have they been incubating?

Please stop giving advice, as you have no idea what you are talking about.

About half of them look contaminated, and the other half are too out of focus to tell.

There is no need to loosen the lid for "better air flow", there are holes in the lid for this.

What makes them look contaminated? I'm not really sure what to look for other than colors that are not white.

I would say most of your jars have a bacterial cantamination of some sort....the mycelium looks stressed out....are you using an incubator? I had a couple jars that looked the same when I was using an incubator

Eek that's no good.

Did you

* Wipe the inner lid after loading the 2:1:1 verm/brf/water?

* Wrap foil over the lids while PCing?

* Flame the spore syringe (no alcohol wipe) between jars?

I used the correct ratio and wiped clean before creating the contam barrier.
I did put foil on them but I think I pressure cooked too intensely and water got into a lot of them.
I did not use a flame on the syringe but I did use a glove box and opened the needle in the box and use alcohol wipes on the syringe and lid of each before each inoc.

Should I trash these? I'm about to inoc another 10 but I am scared about putting them in the same incubation chamber as these now that you all seem to say they are contaminated. My new jars have SFDs and foil so I haven't opened the cooker yet but i'm really hoping they didn't get wet.

A torch or lamp is all you need for the needle until it's red hot. Dispose of a half mL or less to cool the needle. Wiping alcohol is a contamination vector, both on the needle and the lid.

No such thing as "pressure cooking too intensely".

Here is one of your problems, alcohol does not sterilize, but heat does.

All alcohol is going to do is potentially spread around mold spores.

The first drop or 2 cools the needle.

Oh, right. Thanks for clarifying, never been good at small measurements and haven't touched a syringe in 16 years. Nice to see you back BTW I hope all is well.

So should I trash all of these before doing the new ones tonight? Here are some better pictures.

Also if the needle was opened in a glovebox would it still need the flame? I just flame before the first inoc right?
