Ok for bout three months i have had weak and contaminated flushes. One tub would pin super heavy but never grow past the pinning stage, one tub would show green patch and rest of the tub would flush out before being eaten by the green mold.. I have a tub fixing to finish and its got really small fruits so today im taking a fruit from said tub and taking a cut from said fruit to agar. I have read you need to open up the inside of the fruit to take a clean sample.. What i need is a step by step on what to do like 1. Clean SAB 2. Place agar dishes inside. 3. So on so fourth
Many thanks

I know I've read a tek on this somewhere. Have you tried the search at all? There are three of four Teks in that question that you should familiarize yourself with. Try starting here

Best way is to take a biopsy from the inside of a stem and put it on agar. Then do a couple of transfers so you have a fairly clean culture to work with. After you get a good plate put some wedges to a grain jar. Then you can grain to grain etc...

I typically put the fruit into a sealed bag. I clean down my SAB with 70% alcohol and throw like 6 agar dishes, scalpel and lighter and the sealed bag into the SAB. Then I spray the SAB with 99.99% disinfectant and let the room settle for like 10-15minutes.
Go back in, put on gloves and wipe them down with 70% alcohol and begin work in your SAB. Open the sealed bag and take the fruit out. Gently tear it at the base of the stem. Flame sterilize your scalpel red hot before EVERY transfer and cut off a chunk of tissue from inside the stem and transfer it to your dish. Try to make sure you gloves and arms don't hover over any open dishes.

5 out of 6 dishes lived from this clone and they all look great Time to do first round of isolation's. Marked the areas worth continuing.
