invitro poo bags
    #23152058 - 04/25/16 12:31 AM (8 years, 8 months ago)

hey so i made some invitro poo bags , some like at least 2 weeks ago, they are all have been hovering between 80 and 90% completely white, but there are patches of poo that just wont seem to colonize.

im thinking, maybe i didnt mix it up well enough and they are just too chunky? they totally shuld be done right now.

i really wan to try fruiting some to see how well these strains respond to the bags. i could always move them from room temp (75ish) to incubator (82ish) , but like i said they totally shuold ahve been at 100% by now. no contams or nothing, im just not seeing them completely white.

i'll post with pics tomorrow, the substrate/spawn are anywhere from 4-6 inches high, and its kinda tough to make an even surface when doing invitro bags

in the meantime, any thoughts? could i be right that the patches that refuse to colonize are simply just too compact? will that make a huge  diff once i introduce light and drop temp to the 60s?

has this happened to anyone before?

again, will repost with pics tomorrow but any nightowls , feel free to comment

thanks

Doesn't matter how packed a sub is, it will always colonize. Mycelium is mostly microscopic, we only see it because it grows so dense. When I was using millet, my jars always had this giant slab of insanely hard millet the size and shape of the bottom of the jar. It was literally a starchy mess kind of like a semi dried piece of dough but mycelium still colonized it.

Pics speak louder than words, it could very well be colonized only not with thick mycelium which would be fine or something, like bacteria, could be making it stall. IMO you should shake it one last time to see how long it takes to recover, that will surely give show you if something is wrong or not.

well these arent jars, they are already spawned to compost into filterpatch bags

like i said, they usually colonize in ike a week or less in my prior experience but im using a new source of ocompost and i dont think i broke up all the chunks enough

what would happen if i try to fruit at 90% anyways? i COULD like use my fingers to try and break up the chunks with my fingers thru the bag...

def will hhave pics tomorow

im sorry but i didnt keep track of dates

I didn't say you were using jars, you missed my point.

You said that maybe the substrate is too dense for the mycelium to colonize and I explained why that doesn't happen. I have had an inch thick clump of grains in jars which turned out to be impossible to break apart by shaking but since the mycelium is microscopic, it doesn't matter cause it will still colonize it, albeit a bit slower.

If your bags are not colonizing than there is something else that is making them stall, like bacteria, which is usually invisible in subs. When I get jars that won't colonize a spot, I will shake them to see if they recover because if it is bacteria that is stalling them, they will either take a long time to recover or not recover at all. If either happen, I will not use those jars.

well there was no bacteria when i spawned them and the bags look very healthy

theres just random patches of poo that refuse to colonize.  i gottttta get some rest , will repost with pics later . like i said, they've been at like 80-90% id say for at least a week now and I dont wanna sit around and wait.

besides, whats the worst that can happen if i try to fruit the bags anyhow at that high a percentage , esp if you're saying theres probably microscopic myc in the poo and we're going to assume theres no bacteria. i work in a very clean environment and have <1% contam rate. its just been yeaaars

spawned? You mean you colonized spawn and then spawned that to poo inside a bag?

Bacteria is invisible for the most part, we either see them when they get out of control and the jar/bag becomes slimy or smelly or when the mycelium starts producing metabolites. That said, just cause you don't have either doesn't mean there isn't any bacteria. In truth every grow has some bacteria, it is just impossible to have it completely free of it.

i hear you. they smelled fine, no slime, my lids are solidddd

i had 3 jars bacteria out of all the ones i made and they were like 4 weeks old when i re-noc'd them with a new strain cause the first one was bunk. which is totally unsurprising . im going to take pictures of the bags i made , and yeh i meant i spawned them . gimme a few.

thanks dude

So if you are doing invitro poo bags, why not sterilize the poo and inoculate it with spores/live culture? It seems to me you are making all the work of a mono only to spawn in bags.

yes i am doing the work of the mono in a poo bag. its called hydro420's invitro tek, i can link it , each bag produces about .5-1oz if the strain likes the enivironment

i wouldnt sterilize poo mixture because that kills bad bacteria. i just pasteurize... this is how i used to do it and i had great results. problem is, i have no idea how these 2 strains i have going are going to respond in bags.

im going to make some monotubs tomorrow though, and this time i'll use miracle grow moisture control rather than a compost mixture (i had good success in the past using it in trays in a martha, but i dont have the money to build a martha at the moment), and case one of them .. ( i never cased, gretchen taught me its not very necessary and she taught me all i know) .. responds well to the tubs, ill stick with that.

MGMC contains coir and like i said it responds great in a martha, so i assume i can just pasteurize that straight out of walmart and make my tubs. I forgot how many pints to mix in with a 42 ltr tub (one of the small ones)
if neither grow well, i'll try tubs again with the compost mixture ( i have enuff jars to play with, esp if these LC's turned out well. Never had an LC fail on me. IF ONLY I HAD LABELED WHICH STRAIN IS WHICH LOL cause I dont really know what happens when you mix 2 cube strains together except "they fight each other"

i have 4 LC' going of 3 strains (going to inoculate with 7 test jars today, prior to making backup syringes of each in case they contam during noccing), and unfortunately i was too high on dissos to label ANYTHING back when i started this grow so i have no idea what strain is what so im just color coding and hopefully i mix the right strains into the tubs lol

prayinn to god at least one of these strains likes the bags cause they are VERY low maintenance, *rarely* contam until 3rd flush.

chcek out hydro420' invitro tek, its on the forums, or i'll link ya once i take some pictures. just woke up , cofffee time and then ill repost

thanks again

Well, okay. Just saying, it would be very much less work for the same results if you were to sterilize the manure and inject spores or live culture.

Manure needs to be pasteurized only when spawning because the spawn you mix in has to colonize the poo before other stuff grows. When your sub is sterile and you inject a sterile solution, you do not need the beneficial bacteria.

Pasties invitro straw tek follows this logic to a T. I think that tek is still stickied in mush cult so you might want to take a look. You can easily do it with any substrate as long as everything is done with sterility.

Dude I have 4x 5lb Sterelized HPOO/Straw Bags and I just recently completely changed my grow setup and plans and had no use for them. I spent $100 on the 20lbs. Yeah I know I over paid by 10x but I've learned since then.

Anyway. I'm making a bunch of LC from Agar and I literally have 4 Cubes I'm working with so I can just noc those HPOO bags right up with the LC from each and I'll fruit invitro.

Sorry OP. Just thanking him for the help and throwing my hat in on this one a little.

Great thread.

Yeah, the point of invitro fruiting is to reduce the work needed to get fruits and that is the fastest way to go about it.

yeah. sorry been so slow on pics.

anyone know if i'll have luck making simple monotubs with MGMC (contains coir, cheap, i can pasteurize it and then use that to spawn too, rather than a compost mixture)? i was thinking bout making a couple of em, one cased and one uncased.

will have pics anyways tomorrow , but i got a lot of jars colonized that i need to do something with and i dont have the money to get martha supplies right now

Dude honestly your shit doesn't even make sense. I would find a a Tek for each step. Bookmark them. Follow them to the T. And post with questions.

I'm a noob so I might be missing something but I'm pretty sure your adding steps and kinda just making shit up (not like lying, but plans) as you go.

naw i just got adhd. i used to put out about a pound a week lol, its just been years . mainly did invitro bags, but i never had issues with them not 100% colonizing so im thinkin  bout tryin some new forms of bulk substrates

OKay

heres some front/back pictures. the "top" of them are also about 80% give or take colonized.

im going to try to fruit them anyways. i think maybe i just didnt break up the poo enouhg>? i got the compost from a very good source,  but they use it for industrial farming.

as I said im going to try MGMC (in monotubs) because i had great luck last time i grew, but that was in a martha in a tray.

I have a lot of colonized jars, I'm gonna make a couple tubs, but does anyone know if MGMC might work as well invitro poo bags, rather than a compost mixture?

http://imgur.com/a/yIFXh

^^^ heres the pics i took. these bags have been colonizing for like 2 weeks at least. around 77F on average. i dont think they are ggoing anywhere so once i get the temp here down to low 70s/high 60s, im gonn try to fruit anywys. i dont think incubating them will help unless someone else does.

sorry bout my confusing messages

upload your pics via the shroomery, it is not safe for us to press offsite links so most, inculding myself, will refrain from doing so.

What is MGMC? But yes, it is ok to put that substrate in a monotub, you will have to recolonize it a bit again though for it to become a whole again.

sorry, these are the 4 bags , shots from behind/front / some other angle... I also have a couple other bags of a different strain that seem to be colonizing slowly. I may throw them in my incubator ( I know thats a bad idea but I dont know whats going on! Something with the compost, I highly doubt this is a bacterial issue)

i def dont want to open these up and put them in a tub. what im ssaying is, theyv been like this for well over 2 weeks now, they stopped ccolonizing at some point and just wont finiish to 100%. im going to try fruiting them anyways, but since i have  bunch more colonized jars, i'm thinking about using Miracle Grow Moisture Control (MGMC) , it contains coir and lots of other goodies, worked a GREAT in martha trays back when i grew in 2011... any idea if it would work just as well in bags?

I also am going to make a couple monotubs with miracle grow moisture control.

In the past when I made invitro bags, I went to farms, and did everything myself, rather than collecting compost from an industrial mushroom farm. I could also try that again. My theory is the reason they arent fully colonizing is that I simply didnt break up all the clumps well enough.

But yeah I'll try fruiting them anyways because they are clearly not going anywhere, hopefully they will pick up. I dont know. Never had an issue like this.
