Agar plate to grain to manure--can it be done?
    #23036217 - 03/23/16 02:45 AM (8 years, 9 months ago)

I was wondering if it is possible to cultivate some mycelium in an agar plate, transfer it to a bag of rye berries, have that colonize, and then transfer those fully colonized rye berries to a bag of sterilized manure.

How long would the rye berries take to colonize? How long would the manure take to colonize?

I'm working with 2lbs of rye berries and 5lbs of sterilized zebra shit. How would I do this? How do I avoid contamination in the meantime as I wait for the rye berries to colonize?

That would be the proper way to do it yes. Although I dont usually inoculate spawn bags directly with agar I usually inoculate a master jar with my agar wedge and G2G to the spawn bag.

I go agar wedge- grain master jar- G2G to either one or two 5 quart spawn bags. I then spawn my tubs with a 5qt bag each

inoculation, and grain to grain transfer need to be done in a still air box or in front of a laminar flow hood. Spawning to bulk substrate is done in open air, its not a sterile procedure.

You wouldnt sterilize manure based substrate however those need to be pasturized.

I didnt see this part. Avoid contamination of what, youre manure?

Well for one you would just leave your manure around untill its time to use it. Its Poop..its not clean to begin with.
Two, you dont sterilize manure. It needs to be pasturized.. by keeping the poo above 140 but below 170F for 1 hour.

You would thrn mix your coloniZed spawn and your pasturized substrate together in a tray or tub to let colonize and then fruit.
