You don't need to be able to see through it. Your adding food coloring if you follow the tek. What matters is whats on the surface.

Pre-sterilized individually foil wrapped cotton buds to swipe the spore print to agar: is this a good way to streak some spores, anyone?

Is it okay if water is touching the bottom of the containers?
Will this cause condensation issues? Should I elevate the bottom row above the water?
I know you didn't mention it but i'm just mainly wondering how much you use.

Also I'm wondering if the water in the PC should be room temp when I start and slowly bring it up to temp?

I like to elevate it simply to prevent the water from jostling the containers about. This helps ensure they are still level when they cool.

what ratio should I use for that wheat flower? what ammount of potato flakes should I use for good rhizomorphic growth?

Ive made some plates before and they worked well. I never covered tho but thab again i was using baby jars and my own sort of tek with the recipe from this tek. IIm outta baby jars nd dont come across them much so i bought the pp5 containers. Is it kool to just tape it nd pc instead of stuff with paper towel than aluminum foil?

it's gotta be micropore tape. get it at walmart or rite aid in the pharmacy section. no-one is just stuffing their holes with paper towels

^ yep. I had problems finding micropore at first so I used transpore- sometimes it wouldn't survive pcing so I had to reapply, and I had about a 8% contamination rate by using it vs 0 with micropore. (Judging solely on pre-inoculation and agar transfers- not MS or clones)

Waterproof tape, polyfil, and SFD's are also great substitutes.

Pasty did you try a no hole PC run with the lids just slightly cracked and covered in foil

Once done Pc'ing an cool just simply close lids as you remove from PC?

Yeah I did. It worked for the first run but was actually a real pain to get the lids to sit just right. After a first run the lids expand just enough so they don't want to sit cracked in the PC and all got sucked down and crushed. I decided it wasn't worth the effort compared to drilling a hole and slapping some tape on.

I use it because sometimes the heat melts the glue of the micropore tape to the foil. This can tear the tape when unwrapping. I also prefer not having my filter material soaked if at all possible. Finally I like to write notes on it rather than mucking around with masking tape.

I've only had one out of god knows how many get the paper towel wet. If you fold it right it'll lock right in kind of. I like them because they help plates dry faster in SAB and by the time they're done I just get to work. I'd never skip the paper towel as this seems to work just dandy.
