3 Minute Agar Prep - No Pour Tek Pour 1

Everything about Agar

This is MUCH faster than stove-top agar prep
This is the Pour Version of the Microwaves Agar Tek Really all you have to do is:
1) Measure out your ingredients. 2) Add the appropriate amount of water to a quart jar 3) Dump everything in there. 4) Stir with a spoon. 5) Microwave for 1 minute. 6) Stir again 7) Repeat steps 5 and 6 three times and it is ready for the PC. 8) PC for 45 minutes to an hour. 9) Let cool in the PC for around 3 hours. 10) Agar will be ready to pour into petris or whatever you have.

I've boiled ingredients before pc'ing and I've just add them to the water and then pc'd. Haven't noticed a difference.

Oh wow, awesome. I am starting agar today and was planning on doing Pasty's easy agar tek. Wish i wouldve gotten some disposable petri's as well, because I would really like to pour into petri. Looks so beautiful. So with this, instead of PCing the agar already poured into your dish(mini glad thing for me right now), you just PC in a jar, then pour while inside SAB? is that the only difference?

Yes, that is how all pour teks work.
This method actually works better as a no-pour tek. I've found that the microwave distributes heat unevenly, and the bottom of the jar will have thicker agar than the top due to the temperature differences. As a result of this, I prefer to give it a stir before pouring.

What's the point of steps 5 and 6, and repeating? I usually just dump my agar and MEA into a jar, add water to a little over the 500 ml level, give it a quick 2 second stir with a spoon, then put a lid on it and it goes in the PC.... As long as I give it a little swish before I start pouring, every plate is clear and thoroughly mixed.

So, just to be clear, if you plan to pour into petri dishes or something like that, you make agar, put in bottle, PC, then put in SAB while it is still warm? that way it is still liquid? then you pour into petri while in SAB? then what? do you let it cool for a while, then add your spores or cutting or whatever? How many of you prefer the pour method over the no pour? and why? and also vise versa and why?

It definitelely doesn't come out of the PC evenly mixed for me.. Maybe it's because I waited for it to cool. The point of 5 and 6 repeating is that if you microwave a jar with agar for 3 minutes, it will almost certainly boil over and make a mess in your microwave. Doing it one minute at a time allows you to mix and pay attention so you don't have a jello mess..

Yes, after sterilizing, put into SAB to cool, pour while still hot. Once you pour the petris, let them cool and harden before using for inoculations or transfers.

Well because you cant no pour disposable plates. Pitres are much easier to work with, easy to wrap and stack and see into. Not to mention extremely fast and simple to pour. I use no pour when I am out of plates or to save plates.
When pouring, wait untill the agar is so cool it might start to solidify if you wait any longer. You will get a perfect pour, 0 condensation

Thanks guys, appreciate it. So i did pastys tek and I guess my holes in the lid werent big enough, they were 3mm and I am assuming his were 6mm/.25in. So i have lots of condensation now. Are these unusable? or is condensation ok, just making it a little harder to see?
