Advice on cloning please// KSSS w pics
    #22786067 - 01/16/16 11:56 AM (8 years, 2 months ago)

So I've got some orrissa monos going from MS. Roughly 5QTS spawn to roughly 2000g dry of coir/verm/gyp/castings mixture. Next tubs just have coir/verm...

They've been going slow, winter I'm thinkin.

Waiting patiently for em all to form and pop, I've noticed one big guy standing out came up seemingly over night.

I'm still little confused on how genetics work, trying to understand them more so.

Is the big guy in the pic worth cloning lol? Will he possibly carry the genes for big pins?

What kinds of traits do u guys look for? I'm looking for higher BE mainly myself.

From what I understand, cloning a big fruit could lead to big fruits , but it will prob just mean bigger fruits and fewer of them..?

Maybe someone can help me here a little bit

I know the tubs look dry, I misted them after the pics were taken lol. The big guy is in the far left pic. He is bigger than any other pin. The other pics are simply for comparison

Even some of big pins that have been hanging out in other tubs close to his size that formed days ago haven't gotten bigger than him. He's a beast of a pin lol

Don't worry so much about it. Unless you could do a genetics test and reliably correlate that information with phenotype the answer is you'll just have to clone what you like and test the clones out.

I feel you there totally lol. I guess I just gotta go for what I thinks right. I've cloned before, but I waited until they were mature so I could see how they actually flushed ya know, not just how they pinned

I just dunno if I should clone this big guy or not. I'm not looking for big fruits. I'm looking for high BE potent fruits. I don't really want all huge fruits... but maybe that guy is aggressive in some way that'll be appealing to me lol...

This orissa ms culture isn't very aggresive, no rhizomorphic growth I don't really care for how it colonizes bulk. Slow and cottaNY mostly so I Def plan on working on her

Most clone cultures are already significantly better performance than most MS grows so just testing alone is doing way better

when you go to clone that guy, sterilize 1-2 jars of water. then dunk the fruit in the sterile water to help dilute the surface contaminates. 1-2 bathes will increase your likelihood of success. I know most of my contaminates come from trying to clone unclean little pins from my subs.

I've always put pins right on agar. That's the one time I do wear gloves lol. Alcohol wipe hands. Pluck. Plop on agar.

If they're bigger I'll clone them without gloves. Rip fruit in half in your SAB. Flame scalpel cut chunk from inner flesh. Put on dish.

You'll be taking a transfer anyway but I rarely get even a satellite colony on the first plate.

I only do that if the pins come from an agar plate. they are usually really clean from there. I guess its just a lottery thou, and all our experiences will vary with contams. I think, on average, about 50% of my non-bathed pins that come from subs will go bacterial. but I rarely rarely get contams on transfers, so I'm pretty certain its not my technique.

Thanks I was wondering about this

I've got a few tubs going lol so I'll watch all of em and selectmany diff fruits. See which one treats me best.

I'd like to cross orissa w something else, maybe an isolate of mine from a wild print... one day, I'll kept u guys posted lol. Or maybe an orissaxKoh samui to start

Anyways I'll prob wait for fruits to mature, maybe do both... pins and mature fruits.

I thought I recalled h2o2 being used to dunk pins and maybe bleach wasn't sure..

I personally have experimented w cloning from tissue that's been let dry out for a controlled amount of time.(got idea from tradd cotter book )It seems the dry tissue possibly less susceptible to bacteria. I also like to use old plates w a dry surface like a month old to keep bacteria down. The dry plates was an accidental discovery of mine. The two combined led to this

cys from wild u can see where they took over the mold that started to grow lol

Well James isn't looking so peachy anymore. I'm thinkin vert...

I tried using med poly all the way, instead of light one the ends tight on the sides...

I'm thinking I over misted, possibly in combination w having poly to tight all around...

What u guys think? The pins w out caps worry me make me think something else possibly this is a new culture for me not used to seeing that...

Wishing I wouldn't have changed anything now lol...

I'm gonna loosen poly, fan, should I pick off the bacterial/vert looking pins also?

your pins look fine. the pins without caps are mutant blobs. I would let that thing do its thing.

I dunno, I'm almost positive the pins are going south.

They're stalled, real late to pop I've been wondering why and was thinking bacteria...

Now I'm pretty sure it is bacteria. Today they all changed in appearance in a bad way. The pics aren't great tho...

I hope your right tho...

They got fuzzy, the caps got sunken in on the.center of the cap.. it was a sad sad sight this morn I'm telling ya

There is james, as promised.

See how the caps have indented tho? Reminds me of high co2/vert

The dark legions on the fruits make me think bacteria also. It doesn't smell bad... but she was spawned on 1/19... took about 15-19 days for full colonization due to winter temps... I'm awfully worried lol...

I'm thinking that the tubs maybe were just over misted and caused my flush to abort. I dunno. I Def misted lots more than I usually do hoping for better results. Might have crossed the line.

I'm waiting it out, letting em dry out not doing anything yet except watching. They don't look much diff today..

Such for my cloning thread qs lol. I'll get back on track w cloning soon more tubs coming

You don't wear gloves most of the time? What about sleeves?

The pins Def had issues, entire flush aborted. Many theories why for now I'll wait to see.

Still haven't gotten a second flush either.

Meanwhile tubs are pinning that were behind those that went south. So we will see what happens w these. They should give me a lot better idea of what went wrong w the tubs I posted pics of.

I'm not new to tubs have had plenty success, so I know I didn't do anything wrong except maybe/prob overmist.

For now I'll wait for the fungi to tell me what really happened... bummed until then lol

Well it wasn't me it's the genetics.

I've never seen shit like this. I feel like bashing the vendor. They gave me a bad syringe.

Replaced the syringe w two bacteria ridden prints and a new syringe I gave away to member ganja burger for free. Asked him how it treated him no reply yet.

Big bummer bringing back out my old trusty NW clone. :/

First pics are new tubs.

The last two pics are the tubs I posy pics of above awhile ago. No second flush came in  :frown:

Learned a hard lesson here. Always run your trusted shit. Never trust new shit. Never.
