Incubation taking forever...
    #2273635 - 01/26/04 04:39 AM (12 years, 10 months ago)

This is my second experiment. My first experiment went fine: I had one syringe and divided it up between six jars; I kept them somewhat heated and they fully innoculated and pinned in about a month and a half. Since then several syringes have been made and more jars innoculated. This time jars were injected with 3/4's of a small syringe in hopes to speed up the process, which worked. Most of the jars were more than 50% colonized after a week. The jars are somewhat heated at 75 degrees in a dark area--they maybe get light for 30 minutes a day. Some jars have been sitting for more than three months, others are going on their second month, and still, I see no pinning on any of them. They are all completely covered in white mycelium, and some of the "younger" ones have bits of yellowish primordia. I have thought about putting some in a terrarium for a week or so to see what would happen. Has anyone had similar problems? Anyone have any suggestions?

I would never leave a cake in the jar just to get invitro pins. Some people might espouse the benefits of this but I couldn't be bothered. Just get them in the terrarium, into the right fruiting conditions. The jar is not the right fruiting conditions. Get some perlite in the terrarium and I'm sure if the jars are still ok they will fruit. A cake afficionado might want to tell you that you should dunk, I don't know about that because I would never use a cake again. You'd best dunk to make sure they are nicely hydrated.

With the jars being so old be aware that they could have some bad bacteria in the core or they could be extremely susceptable to contamination.

You should really try to incubate at 86 degrees that will help out a ton

I'll increase the temp, throw out the old jars and put the newer ones in the terrarium. With my first experiment they pinned in the jars. Oh well, I'll see what happens. I use jars because it seems like the most simplistic way. What is are some of the best ways in terms of speed and yield?
