Tic Tac Toe Agar (Tiger Drop 2.0) 16

If you have started using agar and playing with my "easy agar tek", and doing the "Tiger Drop", you might be looking at stepping your game up. The Tiger drop is kickass but, sometimes I want more than one massive nock point. The Tic Tac Toe drop is the solution for you. Basically turns a plate from 1 nock point into 9, all it requires is that you have a clean plate, and a sterilized blade.
The procedure is simple. First you need a colonized mini round no pour plate. Then in either a SAB or flowhood stream, take the round, remove the top and with your sterilized blade make 4 cuts in the agar, in a tic tac toe pattern. Go from edge to edge. Then perform the Tiger Drop with the plate.
The agar is now in 9 still pretty significant chunks. You should see very rapid colonization, especially good for slower colonizers. I like to shake it up real good still. Get the wedges dispersed.
Half Gallons with tic tac toe

Shake em and spread em out a bit

Sometimes a wedge or two may fall off the rim. This is no biggie, just let it go. 7 good size chunks will still colonize pretty fast.
I like to use this for my half gallon jars and spawn em to minis. One culture one jar one sub. Great for quick testing and not needing to expand. They are also really good to go with to pint masters. A pint master with this will colonize very quickly.
I will be fleshing this out some more so consider this a work in progress. . .

Oh ok! I see now. Thanks for clearing that up.
I can see the advantages when compared to cutting the media in a pie fashion. Greater surface area, equally sized wedges, && I bet the squares don't fall apart and break during transfer like the pie cuts do.  Ive always favored cutting my wedges like squares rather than a wedge.

Why not a * pattern that centered on your growth? Just seems like you could cut that into more than 9 segments and have many more noc points if you're going to bring a scalpel vector to the game.

Cut it into as many as you like, but straight lines are quick and easy to cut, and 9 wedges are plenty, so I cut it this way. I can make 4 cuts in about 2 seconds and tiger drop with ease allowing for minimal exposure. I guarantee that no one using petris can drop 9 wedges with a single motion.

You have just inspired me to try this with the couple pasty plates I still have chilling in the fridge. I dropped an APE wedge on a couple quarts last night, but I'm anxious to try those little fuckers, would be awesome to get a couple quarts colonized *fast*.

I like it. I do this too, except I just fork all the squares on my scalpel blade and drop them in at once or in 2 drops.
A full plate in a pint master is way fast!
If Im just innoculating normal quarts I just drop a 1/4 wedge in each jar

Slower leap off. Longer exposure of the agar media. Larger wedges can rub myc off onto grains when shaken. You could take the time to cut the plate into dozens of bits, it won't colonize any faster but you do run a greater exposure vector.

I fully endorse this as a second edition of the tiger drop.
This one requires ninja precision. It requires preperation. Most of all it requires smooth and deliberate technique without fear of failure.
If you hesitate during the cuts you risk longer open-exposure.
This is the kind of mycology they train for at the shaolin temple.

I was suggesting the use of laser pens to kill contaminants on agar before, some said this could just spread them as it vapourises. But maybe a laser could cut up the agar without the need to open it. I don't have a laser pen so cannot try it.
I was also suggesting having a sort of grid made of metal that could be pressed into agar to chop it into lots of small squares all at once. Like things for cutting up potato.

I fucking hate punching 6-wedges. Seems so lazy to not just put the extra blade in to make 8 that come out more even for cooking.
But a blade like that could be fun to use. Maybe I could make like a 'drop machine' that could be sterilized and attached to any miniround inside an SAB and then you manually turn it over onto the grain jar.
Instantly a vacuum will suck the puck out and pull it through a hot six bladed hole. Then the bottom opens briefly and the pieces fall through.

I've had this dream lately. Robotic SAB.....
what if the plate had a grid. Think like an over-filled ice cube tray. Could it colonize entirely from the center? Or a full swab rub? It would hopefully produce an almost perforated puck that could maybe be broken up during the shake....?

Sorry to bump this older post, but I have to point out that this minir improvement over the original Tiger Drop is exactly the extra step I needed. Tiger drops always colonize slow, stall, or wait to colonize and draw from the agar until it's spent of nutes.
I'm going back to agar and doing this with all of my extra plates. Thanks for sharing the good idea, Pasty. (Pods are still going great, too, btw)

I've been reading non-stop on agar related teks, and I'm interested to know if there's been any concern in cutting the plates like this. Obviously it's working like a charm, but I recall RR mentioning that cutting the myce can harm it in some ways like general health and growth (vague—can't find the quote now).
Don't mean to bump this thread on such a neutral thought, just wondering if anyone can attest.

How are you supposed to make transfers to clean up cultures and isolate without cutting the mycelium? RR would never say that. At least I don't think he would, perhaps you read it out of context.
But it does work pretty good.

Mycelium can even withstand and recover after the onslaught of being blendered, which it is done to make a LI. The tek in the OP would be but a gentle caress compared to the above-mentioned tek.

i like this tek more and more....went to g2g a regular tiger drop and the wedge more or less got in my way, not an issue when i use this method

Little testament to the results of the 2.0 vs the OG tiger drop 
Both jars were started from KSSS plates 5 days ago. First is a plain tiger drop, the second I cut into roughly 9 or so pieces and shook up in the jar. They are both colonizing beautifully, but 2.0 is just about finishes while OG is a little behind. OG-

When i wrote the tiger drop tek in a tiny little coffee shop in BFE i thought no one would take it seriously. Now it's been improved and people are realizing how perfect a delivery system the high-walled petri dish can be

2 healthy pasty plates.. Im so nervous on using a scalpel. How do I cool the scalpel? Wouldn't dabbing it in a sterile agar plate, then cutting into the agar contam it?

I go in hot too. It might hurt leap off a touch but with these you get so much inoculation power it hardly matters.

I just want to say that when I made my first "G2A to A2G" isolated transfer of Penis Envy with a "pasty plate" (using the easy agar tek) and made the Tic Tac Toe pattern in the agar and popped it into a quart plastic bottle with grass seed (using the pods tek), this thing became 50% colonized in 5 days! I thought G2G was fast but this was magical. It looks so powerful, too. Anyway. Thank you for all for the great info and easy learning.

Pasty. Your plate tek is the reason I'm still in this hobby. I didn't realize how feasible agar work is with widely available PP5 containers. Before agar I always had mixed success. Come to find out all my syringes and prints were dirty before I ever got my hands on them

Can this process be used if I'm using a spore syringe to inoculate the agar? I know your original tek using the Tiger Drop was for use with a print.... as long as it's not contaminated is that important?

Your spores are contaminated. All spores are contaminated. You must transfer them on agar 1-2 times to clean the culture before using this tek. It doesn't matter in the slightest if its a print or a syringe. The spores dont know and dont care.

I've got a clean lookin plate grown to the edges and some old jars of grain that I tried to inoculate but my wedge was so small and thin it didn't take, but they look the same as they did out of the PC. I'll be gone for 3 days, who wants to wager what I come home to hah

Yea I just figured u might have some insight on how to do it without getting anything contaminated. Since it won’t be just a simple squeeze the plastic and have the agar puck fall in. I’m gona need to like set everything down and scrape off the pieces into the jar which is gona suck And open me up to a bunch of possible contams. Welp here’s to hoping it works out.

You will be fine. Just flame sterilize your blade, work in a SAB or in front of a flow hood. I cut mine up like pizza, from one side straight to the other until I have slices and then try to squeeze them out, but most of the time they stick so I use the scalpel to nudge or drop them into the jar. I use both pasty plates and the ziploc twist n lock minis. The ziploc container is firm and hard to squeeze so its a slice, scrape, and drop game depending on the containers im using. Dont over think it, you got this!

Just did this but cut a circle around the tic tac toe portion instead of cutting to the sidewalls. Black agar in the grain now.
