I've never changed my surgical tape on the plates, keep using the same paper towels, lids are super loose, etc. My contam rates have not gone up.

I don't even use the paper towels anymore :shrug:

I think we had this discussion in this thread already between me, Muda and a few others. We all re-use the same foil, don't wash our plates, don't use paper towel and don't change our micropore.

Pretty much set-and-forget tek :thumbup:

Yea dude at first I was like "man these lids are kinda tight" then "man these are super loose did I fuck them up" and now Im like "man I kinda like them better loose it was too clumbsy before" :lol:

Gotta remember not to pick them up by the lid though.

...Uninoculated plates got contaminated from (I suppose)adjacent contaminated plates(forgot to get rid of)... micropore doesn't provide enough barrier for such a situation... have you guys any experience with this or is it just me? I'll stuff the holes with a small polyfill next time around..

It's never been a problem for me. Moste of my plate have poly-fill or SFD on them now though.

I just got a pressure cooker today--can't wait to get serious with this. Does micropore only keep the glad rounds from being smushed? Would 2 layers limit gas flow, or am I being paranoid? Also, I dug up a first aid kit that has what looks like micropore tape. Would that pop up in one of those kits?

yep that would "pop up" in one of those lol, it's there because the hole stops a vacuum from forming so the hole needs to be filtered, 2 layers won't be a huge issue

I believe I asked Pasty about that last time because I had garlic flakes. IIRC, yes, it does matter, you want regular, plain potato flakes. Otherwise there's too much salt in the mixture.

I've grown lots of different plants with average to good success rate, but I'm a total newbie at making my own mushrooms.

Always wanted to do agar. Now I've got 8 containers (Pastywhyte style) in the fridge and one ready-made growbox is fruiting. How do I go about cloning? Just drop a piece of mushroom over the agar? And which tek would you advise from then onwards? There are so many I'm baffled.

Open it up with your hands and then take a transfer inside, don't cut straight into the thing with a flamed scalpel because you'll push contams in. The theory is the outside of the mushroom, which was fruited in open air, will be covered with contams while the inside will not.

After being intimidated for way too long I started some pastyplates! Having a hard time getting anything to start by using a drop from a syringe even though I know it's viable. Other plates either have a colonised grain in each one or a small piece of tissue cloned...all of those are rocking! :awedance:

So what am I looking for to transfer with? Faster rhizo growth? Not getting much of that but it is all spreading nicely. When transferring is it a problem by cutting down into the agar to cut out the myc? And last...is 2 transfers enough to consider it an isolate?

Oh...and if going to store plates in the fridge do you want the entire surface colonized before storing?

Rhizo is overrated.
In fact everything is overrated until fruiting potential is locked down. I whip up nutrient heavy agar. A tad heavier then Pasty's formula, which is already a little high btw, and this favors tomentose growth. All of my agar is tomentose and my tubs come out great. I prefer the slower growth, it gives me time to remember they're there :lmafo:

In that first wedge, the top most from like 9 o'clock to 3 o'clock, I'd transfer from closer to three o'clock, see the quicker genetics expanding out faster then the other sides?

That's isolating for speed, the only thing we can see on agar.

just transfer the fastest growing areas. being rhizo or not doesn't really matter for anything, just looks.

cut into the agar. this is why we refer to agar transfers as "agar wedges" because the mycelium will recoup faster on the established agar than having to find a new food source.

2 transfers will probably make it clean. 2 transfers is generally not enough to find a monoculture. Some people have done up to a dozen transfers before finding the monoculture.

Do you need to find a monoculture? No.

Can you grow buttloads of mushrooms with multispore grows and clone grows? Yes.

Since you are new, I would recommend not thinking about isolates. Like I mentioned, you may have to do a lot of transfers to get to the monoculture, and even then its not guaranteed to even grow anything. So not only do you have to make a bunch of transfers, you have to test all those transfers for viability. Its fucking lengthy, grueling process.

Well yeah the goal was to start trying to get some isolate or monocultures in order to achieve consistent flushes and potency. I was under the impression 3 transfers should result in an isolate. IMO no real point in agar work if not working towards an isolate....may as well stick with G2G transfers if it's MS. :shrug: All I know is I'm pleased the first agar attempts have gone well. It's not a variety I plan to keep around so it's been more of practice. Just one of those things I've been too scared to even attempt...out of the 8 or so I've made no contams that I can see. :zaphod:
