do you let it cool down first using agar?

Once you have your warm agar pressure cooked and poured into your pertri dishes, do you have to wait for the agar to completly cool down before putting in some spores? Thanks!

Well, it has to cool down sufficiently to solidify...I would think you would want it to return to room temp (I always do...). BTW, don't pour the agar too hot or you will have a condensation problem on the petri lids...

So do you want the agar to solidify before adding the spores? Or just when its room temp?

Yeah, the agar will solidify before reaching room temperature...you could presumably streak plate as soon as agar is "solid"...properly prepared agar sets with an almost "rubbery" texture (I have observed), not a soft jelly-like consistency. Agar should not cling to the loop or knife...I find that a sign of improperly prepared agar or not sufficiently cooled agar. Just my opinion...

hmm -- you may want to research more about working with agar/making plates and innoculation of spores into any substrate -- it may make it easier on you when you make your plates.
