Most Oster blenders are recommended, you need one with a specific blade assembly, I'm on my phone atm but if you search it there should be a tek written up about it.

Oster blenders work best, you can find a blade assembly on amazon and attach it to a regular mouth 1/2 pint jar using rtv silicone from AutoZone, the gasket sealer. Then sterilize water for 30 minutes, tiger drop it into the jar in an SAB and blend it. Then pour it into your jars. I do two jars of LI and then G2G those to more qts.

I have worked with broths, liquid cultures, and solid media quite a bit over the years, but never for cultivation purposes and never with mycelium. I would frequently scrape a colony of bacteria with a inoculation loop and make transfers to solid and liquid medias, but have noticed that there is no mention of this for cult purposes.

Is this because there is concern that scraping make damage the network or?

I do that all the time, I also explained LI better up there.

Define your use of solid and liquid media, as in what components and what's lacking in mush cult?

Mycelium is pretty tough, tougher then we give it credit for but just as fragile as we fear. If cut from its original source it'll recover over the span of a day or two and continue expanding.

I'm not talking about cutting a wedge for transfer from the mycelium on agar. I'm talking about taking an inoculation loop to a section on the hyphael ring and scrapping off some of the mycelium either into a liquid or on a solid medium. If we could successfully transfer without the agar, I am wondering if it is possible to transfer it into a prepared solution for inoculation or if that would just destroy the network and make it impossible for recovery?

Ahh,

Absolutely possible :super:,

It would recover. Like I said, tougher then we think but get it to dry, to cold, to hot and you lose half your substrate or inoculant. You could transfer that way into any medium you choose but I'd give it an agar to agar transfer to start to minimize shock and test the theory. Then move on to LIs and such.

Part of the reason we cut into the agar is just for something to hold while we transfer via scalpel. Sometimes the mycelium digs in underneath that initial level of agar though so scraping may prove problematic, especially with drier plates.

As far as liquid media-- we used everything from a nutritive broth (dextrose stock or glycerol stock) to non-nutritive saline solutions to a variety of solid media. I assume that one could not just add the mycelium to water then use it immediately eliminating the need for nutritive component since storage wouldn't be necessary? Or does the mycelium need the sugar in the agar that is blended up in order to recovery?

Mycelium doesn't need the sugar from.the agar to say to recover, if put into a liquid culture it will actually absorb nutrients through its cell walls along with the oxygen necessary for its reproduction.

So if you dropped it in dextrose stock I'd imagine it would multiply similar to dextrose agar but I'm not 100% sure what's in your dextrose stock but if bacteria can expand, good chance myc can :thumbup:

I am definitely thinking of trying out some different LI ideas. I feel like while blending up the agar in water then injecting it into grain might not be necessary if I can transfer mycelium directly into a solution instead-- Hopefully by doing so, reducing the chances of contaminants. The question is what kind of solution/broth to prepare.

OR transfer agar to syringe, add water and agitate to a certain degree, then inoculate immediately?

The reason we blend it is to create hundreds of thousands of inoculation points. Without blending it you'd end up dumping a big piece of mycelium in a jar more similar to a tiger drop and the nutrient water may actually be an added vector of contam.

You could transfer to a liquid medium, shake it to break up the myc and suck it into a syringe but that adds two unnecessary steps where something could potentially ruin your grain jars.

You'd have to find a way to get it finely grounded in the medium to effectively use LI, otherwise you risk not taking advantage of the speed garnered from those tint pieces of agar with mtceliun holding on for dear life :lmafo:

I see. I'll mess around with some ideas and see what happens.

However, my tipped substrate failed. It fell and cracked. It's pinning but is so sad and ugly looking. The Argentina tub is moving into second flush. Looks good.

Well the side that fell facing upwards, started up. It flushed then I flipped the substrate. After a few days I noticed a shit load of pins on the underside and none on the top, so I put it on a rack and tilted it. Now the bottom side is up and there are a bunch of maturing pins that a smushed. Lol. A few are now growing up but many are growing sideways. I fucked with it too much.

Grab a pic before you toss it out. I had three subs I had just dunked and I had them in my living room because I was being lazy and my cats tipped them over and all of the subs cracked in a million places.

I was so backed up with spawn that it wasn't terrible but I almost killed them both that night :laugh2:
