Grain and Agar ?'s
    #2248107 - 01/17/04 12:53 PM (14 years, 4 months ago)

I recently sterilized (using a Pressure Cooker) a number of PF tek cakes, and some WBS jars(fitted with polyfill lids) using the measurements and teks found here, I just started my agar dishes and was worried that my substrates (the BRF and WBS) would dry out...what is the typical shelf life of these, are my concerns legitimate, or unfounded? Also about how long should my petri dishes take to colonize (used a spore print and am keeping them at 80-85 deg f) thanks alot for any help

in my experience it can take the spores to germinate from 10 days to one month depending on strain, temperature, age of spores etc. If you have sterilized you jars with WBS properly you can keep them for quite a time. Just make sure that no water can evaporate and no contams can get in. I would put saran wrap around them and put them in the fridge.

AFOAF had a some B+ sitting around so he decided to do a smear on some PDA. the spores germinated in 2 days and the plate was colonized in 10 days. and they say B+ is a slow colonizer pfffft. of course variabls differ and it may take longer.
