No pour / no condensation?

For the time being I am attempting a no pour useing mini rounds. I would like to move on to Petri / pour but waiting on the Petri's to come in the mail.


So my question: what is the cause of condensation when it comes to the no pour tek?

  I have been doing some reading and yet to come to a conclusive answer. Does the condensation stem from pouring while the agar is still too hot?
Does it come from raising or lowering the PC heat too quickly?

I have been doing as pastywhite has and starting with a mini round, folded paper towel, foil with little luck. Thanks.

Drop the pressure and heat slow. Let cool off overnight. Give 2 to 3 days and 90% of condensation dissipates. At any rate condensation is not a contam vector. People worry too much. FYI pour agar can be a real bitch for condensation. Be sure to use the mug of hot water on the top of your stack trick.

Probably both.

Try letting the agar cool in the mini-rounds before PC'ing and bring the pressure up slowly like pasty said.

I did let the agar cool before "pouring" (I use a syringe 10ml/mini) I will let the temp drop slower this time. Last attempt the condensation stayed for a bit longer than a week and I felt it was not allowing my spires to grab hold to germinate because the amount of water sloshing around. Thanks for the tek BTW +5 pasty.

once you find a nice stable temp they go away fast, the coffee thing is simple, stack a couple pasty plates  or a few petri dishes and tehn put a warm cup of coffee or water on top

Violet pm'd me a cool idea about putting the plates upside down.  If you use filters, they'd need to be on the side or not at the lowest area on the lid.

Haven't tested it but it sounds like an excellent idea to avoid pooling and dump the water asap.

I spawned some popcorn casings and had double-overlay cause I didn't put enough hydrogen peroxide in my automated aquarium mister.  I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave.  I think it will be good cause B+ is so potent.
Triggered yet?

I don't recall having issues with germination on PDA. Really i have had few issues with germination on any media. My preference is no pour cause I'm lazy, poor, and don't have a flow hood. But sometimes i do pour agar for certain projects. When it comes to agar what matters most is that you're doing it. Doesn't matter how ya skin that cat otherwise.

So as far as storing the mini rounds, do you keep the foil and paper towel on top until use? Right now i have them in a giant zip-lock in a storage-tub in another storage tub (just the way it worked out , no special reason) but with the foil and the paper towel still on.
I know the condensation  isnt a real issue or vector for contaminates BUT doesn't mean i don't want it gone =D

I poured the agar when it was close to room temp and covered them them with the paper towel and foil with no condensation, raised the PC temp slowly, 15psi 45minutes, lowerd temp slowly, let settle over night, opened PC, stored plates away till use,2 days later still as much condensation.

QUESTION: my AA921 seems to always have (negative?) pressure when it cools down, any way to stop this?

Ive found the best way to avoid condensation when pouring is to wait longer before pouring. I used to pour as soon as I could handle the bottle without protection but now I give it quite a while longer. I get a couple drops on the top couple plates but nothing serious, my plates have never been better

So once you start pouring if you are having condensation let your bottle cool a whole lot longer :thumbup:

Ah yes. Thanks. Definitely going to get a temp gun and find the optimal temp for pouring once it comes to that. for the time being im wondering about storage, would removing the foil and paper towel be a con? as in allowing spores and contams to rest on the surface. or does it not matter... or does removing it not matter and actually helps dissipate the condensation.

wondering if silica packets or such would aid in removing condensation through the micro-pour tape on the lid.

Some say no pour makes more condensation. I found that plastic petris make less condensation for me for reasons I have yet to understand. Maybe the vents I dunno.

So a better insulator results in less condensation? I knew I thought I saw less condensation this time. And last time I used glass ones the agar was barely liquid. There werent any lumps but the outer edge all around the bottle was set. Still had a crapload of condensation.

Store em in a plastic lunch box in the fridge to avoid temp swings which cause condensation.

Aren't you supposed to store plates in the fridge when you want to slow down growth?

I am apprehensive about using our fridge because everyone uses it and its always full. Can't begin to Imagine how much contams it has lurking around. I was looking to buy a small beer fridge to put in my room before I spent every penny on equipment. At least its at the top of my wish list now lmao

Yeah. Do you mean while the culture is growing? In that case I don't know man my condensation goes away a few days after I make them. If it doesn't and the rounds have some water in them I just dump it out in my SAB before knocking them up.

Get a separate fridge for myco stuff. Your regular fridge is disgusting.

Usually mine goes away while I put them in the incubator. Then when I inoculate I just wipe lids with alcohol soaked paper towel. When I have a shitload of condensation and I don't store them upside down it usually gets bacterial. Haven't been having that much problems lately though. I guess you get used to dealing with it
