peroxide and millet

how would it be possible to have peroxidated millet? if H2O2 breaks down at about 140 degrees F...can this even be done? i'm hoping that it can because it would greatly improve chances of success IMHO. could you squirt some h2o2 into the grain after pc'ing and shake it around? would this have any effect?

well i don't think you'd want peroxide in your jars when you're innoculating them. but if you didn't know already peroxide is light sensitive, it will break down into water and oxygen when exposed to light. so if contamination is your main concern, i would say go ahead and try shooting some h202 in the jars and then leave them in the light for a day or two so the peroxide breaks down. if it worked i'd say you'd have pretty sterile jars. i dunno though, never tried it, but i'd be interested to hear how it goes.

thats the point. but you can't innoc with spore water. Peroxide KILLS spores! If a little H2O2 was squirted in in and shaken prior to innoculation with 2-4 cc of dense liquid myc, or a piece of sterile already colonized myc this would work.

i know that it would kill spores, that's the point. i would be innoculating with a liquid culture...i just want to greatly improve chances of success, that's all. sorta like using peroxide in agar. i was just wondering if anyone had ever tried anything similar with grains.

I dida search for peroxide. turned up some usefull info.

A quart of birdseed spawn is selected from several prepared in the following manner. Birdseed rinsed clean several times in cold water (which btw removes a lot of sunflower seeds if you aren?t careful, or want it to, sunflower seeds float and are easily poured off from the rest of the BS). Boil an excess of water first. Reduce heat once boiling. Seed is simmered on low for 30 minutes (no more since we will be adding peroxide later and don?t want too much moisture). The grain is strained, rinsed and dried thoroughly before being loaded into the quart jar, ? full. The lid has a filter disc in it, but polyfill, or even coffee filters or cardboard can be substituted since we will be adding peroxide later. The jar is pressure cooked to sterilize and also destroy peroxide decomposing enzymes. When cool, the quart is injected with 12 cc?s peroxide, shaken and inoculated in front of a flow hood with a piece of the peroxidated agar. Colonization is complete in a week with temps around 80 deg F. A huge filter patch bag is prepared with birdseed as above. Peroxide amount is proportional to that listed above. In the flow hood, the bag is inoculated with the colonized quart jar and sealed with an impulse sealer. The bag is colonized in 11 days at room temps.

adding peroxide afterwards is very beneficial with contam rates.
peroxide is not broken down by light it is broken down by UV light, leaving it in your room under a lightbulb isn't going to do anything, just a clarification.

btw.if it's broken down by light nobody would have peroxide in the world probably because of the bottle it's in.;)

I thought that was why the bottle was brown? To keep out light!

i just wanted to thank you for that little exerpt...i was originally going to soak the WBS in a peroxide/water mixture, but then remember that peroxide decomposes at higher temps...i'm going to have to give this as shot when my liquid innoc is all ready to go...thanx a lot man!
