How do these plates look?

Greetings all

These are the first transfers I've ever made.  They came from one MS plate.  I can see some contams but it's hard for my noob eyes to tell what is and what isn't.  Can y'all professionals help me out?

I would pull transfers from 1,3,and 5...transfer a couple pieced to new plates...pieces about a quarter of the size of the original transfer...that strong growth on 4 also looks promising.

Do you think 2 looks ok?  Does the agar just two tone like that sometimes?

picture 1, 2 and 4 looks fucked with bacteria. Just make a few transfers. Watch your sterile procedures in the SAB.

Not completely fucked he could transfer from those other plates, but #2 has no chance.

Agreed, 1,3&5 are the best looking, but you can pull healthy mycelium from all the plates. I would get a SAB and a bunch of fresh agar dishes, transfer 3-1-5-4-2 in that order with transfers the size of a grain of rice. They look great though, just make sure to use good sterile tek.

Yea the second plate has that sneaky bacteria that covers the whole plate with a film so its hard to know if its there.

Was there a lot of bacteria on the first plate (that these transfers are from)? If you have a photo of it maybe we can help you figure out why you have so much bacteria on these

Seemed fine to me.  This is what it looks like now.  I don't have any photos of it when i inoculated.

There is a lot of condensation on this plate.  Could that be making it look that way?  The myc seemed to cover the plate quickly

Maybe I'm just in denial.  lol

Did you inoculate with a spore syringe? The liquid kinda slides around and if it germinates everywhere it can grow over mold or bacteria. Same as if you let the germination plate grow out big before you transfer, its the most likely plate to be dirty.

Next time drop spore solution onto a sterile cotton swab and swipe a little spot on the agar. Or a flame sterilized loop.

Take your first transfer as soon as you see healthy growth as far away from contams as you c an get it, maybe wait till its grown out about the size of a dime or so.

You only need 1 transfer from the first plate anyways, you wont see any sectoring for at least 3 or 4 transfers, usually like 8 or 9 to see prominent sectoring IME.

But from the first couple plates you are just cleaning up the culture, so just take a transfer from the healthiest looking spot :thumbup:

Oh me too dude! I mean he only really needs to taket one wedge from the germination plate instead of starting like 5 plates
