Liquid Culture. Can all liquid be used?

Hi All,
I've searched but couldn't find an answer so apologies if it's been asked before.
Can all liquid be used from a liquid culture or only the strands of mycelium? Obviously the blob of mycelium will be actual mycelium but I was wondering if the rest of the liquid contains mycelium as well?
Also out of a liquid culture what % should the expected volume of mycelium be compared to liquid. it seems that mine so far are very little compared to the massive amounts i've seen in some pics here.

If shaken well, 1 drop is all you need at least for agar.

Just siphon out the strands of mycelium for much faster growth. The surrounding liquid may have tiny particles of mycelium floating around, but I don't see a point in using that to inject your jars. Using too much liquid will only throw off the moisture content of your grains and will slow down colonization.

I like to stir it up and aspirate immediately before things can settle, like Psilo said the clearish looking liquid may have myc but it takes a lot longer to recover.

I stir mine up to make sure its broken up, but I then usually let it settle. I then tilt the jar and draw off from the settled area. This can be difficult for some using filters.
If you keep the filter to one side of the cap and have a SHIP on the other side it can be done. I also have jars with SHIPs in holes drilled in the glass at the bottom. The current LC I have going simply has no filter at all. I have said before I think the filters are overrated on LCs as it often prevents you shaking it well, which will aerate it as well as break up myc. If you have a large jar which is only 1/4 or 1/8th full then it has a fair amount of head space in it. The larger volume of air also reduced the problems of vacuums when pulling liquid from a sealed jar. There are other ways around the vacuum problem too, like injecting a full 10ml syringe of sterile water into the jar before sucking up 10ml of LC, so you have equalised it. Or if you have sucked out a syringe full and injected it you can heat your needle and slowly draw in air into the syringe through a flame in front of the needle, now inject this air into the jar and suck up more liquid.
If you have no filters and a SHIP in the lid you can allow the LC to settle upside down, and put the needle in and draw from the dense settled myc layer.
If you are stirring it up before sucking it up you can still concentrate your myc. Simply suck it up and cap the syringe and let it settle with the needle facing upwards. Now the myc will settle against the plunger and you can inject off the liquid layer over a sink or outside. So you might reduce a 10ml syringe to 3ml of denser myc laiden liquid.

I would like to do it your way blackout but my issue with it is:
Usually when I stir up the myc, there are a few chunks that don't really break up that well, and those want to settle to the bottom the quickest. I have issues with the needle getting clogged on that bottom soupy stuff. So my way... I get all the stuff stirred up and a few seconds later tilt and aspire from the liquid very near the surface, which has a greatly reduced chance of having a needle clogging clump in it. I'd love to do it your way but .... maybe I'm not stirring for long enough? I only generally stir for 1-2 minutes.
I can usually get 85 -90 % aspirated and the remaining really thick chunky stuff I end up tossing out..

I often had to suck and squirt out again, this seems to tear it up, but I did get clogged needles at times, I have sharpened the ends of my needle before to help cut it up. I have been using very weak LCs after seeing it recommended by Workman, he was saying just 0.1-0.5%. This usually results in myc spreading out into thinner strands, just like it would on weak agar and I find it much easier to suck up. He also reckoned it might suit long term storage better since there are going to be less waste products per ml and its closer to it being stored in just water and so less stress on the myc.

Sound like a good plan! I've converted to those concentrations recently, just saw the first sign of recovery this morning....
