So I finally got the contams out of my plates and am getting some rhizo growth..it took a long time and i contribute it to un-sterile processes i was doing.

Yes agar will teach you proper technique. That is why its pushed hard on noobs. Once you can get clean transfers everytime, you have the proper movements down for things like LC which, when inoculated with a clean agar wedge, performs killer 99% of the time.

Awesome write up Pasty! It's perfect for people like myself who have never worked with agar before. One question tho, and I apologize if it was already asked before, but would the recipe differ if I am using agar flakes (not powder) instead? That's the only kind of agar I was able to find after going to 3 different Asian markets today lol

Question about transferring to new plates.

Is it better to cut small pieces of mycelium covered agar and transfer or would it be better to scrape some mycelium off of the agar and just transfer the scrapping.

cut wedges. it will recover on the wedge faster then it would on the plate.

Waited til the agar left stuck in the pot cooled, peeled it all off, and put it in a pasty plate with a few drops of water, came out just fine.  Gonna start doing that instead of rinsing it away.

I did my first pasty plates today! Normally Ijust use 1/4 pint masons when I run out of plates but went this way this time.

Although I mixed my agar bottle up first and cooked it for a few min, then poured in open air when it was nice and cool and then put thru the pc cycle.

They came out nice but slanted :facepalm: I didn't weigh them down enough. Ran them thru again.

Hehe pasty slants! This time I put a trivet over the first layer of plates and set a jar on it to weigh it down slightly.

Man I wanna do a tek with a catchy name, acheive  internet fame! :lol:

Leaf have you done any of these with the stop cock in yet? I brought it up to 15 psi only but it was tricky bringing the pressure up slow enough to keep the condensation down. Well see how I did I let it cool overnight :shrug:

no, i just do pour agar now.  i used the pasty plates to make brf recently though :lol:

i love the stop cock dude.  every time the needles goes into the caution zone i pee a little :crazy:

You arent supposed to wrap these things right?

The only problem I had using them, besides the lids and deep bowls being a bit cumbersome.. thats to be expected though.. was loose lids.

Some of the lids didn't really snap together anymore, some were loose enough to fall off but most just arent snapping super tight anymore. I didnt go above 15 psi I wonder what gives?

Im just being carefull moving them around I think all the plates I inoculated were ok, I used maybe 20,

I have like 1-2 pasty plates where the lid doesn't hold on as much. Had a mini heart attack today with a plate that has some APE spores swabbed on. The lid almost came right off :eek:

As for the slanting of the agar, since I have a small PC, I get that for most jars if I let them cool in the PC. Whenever the pressure is back to normal, I'll wait until the gladware is cool enough for me to pick them up, but still warm where the agar is liquid.

Then I just let them cool on a counter or in my SAB until they're hard. If I ever get condensation, it seems to go away soon.

Moste of my pasty plates have been PCed many many many many times. None of the lids snap on tightly anymore. I actually prefer it like that. It makes them easier to work with in my SAB. I also like the high walls. They make it easy to pick them up without getting my hands close to the opening. I just hold them sideways to take transfers from them.

Well that puts my mind at ease some. They'll take a little getting used to but I will definitely be using these over 1/4 pint masons for agar plates when I run out of pitres.

Or even for the first bunch of transfers for isolation to save money on plates!


So even with the lids a little loose they dont need to be wrapped yea?

I never wrap them. I store them in a tote that I periodically spree with lysol to keep there exposure to bacteria minimal.


I've stored uninoculated plates in this thing for more than 3 months and they still worked fine without any sign of contamination.
