A better strain conservation method?

Hi all,

I have some strains which I would like to save for years to come, (Mex-A, Tampanensis and Cubes) and I'm trying to figure out the best way to do so.

I thought about preparing some slants with agar and some wood, but the I read a comment by RR saying that a jar with colonised grains might last longer.

Also, I read that some strains cannot survive the low temperatures of a home fridge (the only fridge I currently have access to)

Any advice on this?

Some "Strains" can't survive it?
If it's cubensis it should survive it as long as it never frosts over.  I believe tamps and mex-a are the same.  Just don't let your fridge go under 35.

Slant some, keep a few colonized jars, and take some prints and swabs.

Slants seem easier, since colonized jars have the possibility of having endospores that fuck your culture.

Slants are great for long term storage. May want to make new ones once a year.
Also, spores stay viable for a LONNNGGG time

if you just want to store the strain a properly made/stored spore print will last for several years  how long are you talking? Ive seen people on here use 7-10 year old prints without issue:shrug:

Well, the thing is that I've worked so hard (and spent so many petri dishes) on isolating and testing killer strains, that it's kind of depressing for me to even think about going back to spores instead of starting off the next "seasons" with some agar / LC of great mycelium.

then you want to store a "culture" not a strain do some slants then I guess ive also heard of people using sterile water for long term preservation but I personally don't know shit about it  :shrug:

Would you recommend doing the slants the way RR is doing it in his video, with a wooden stick inside?

Yeah half a popsicle stick or tongue depressor works great.  It's not necessary but the wood helps with longevity.
