How big was the piece of tissue? All of your plates look like garbage. Sure you could work from one of them but it's not worth the time. Just start over. As for using spore syringes, don't use a drop. Use less than a drop. Press the plunger so that a small ball of liquid starts to form at the end of the needle. Then touch it to the agar. Maybe do what pasty said and make a divot and let the drop go in there.
If you must use your current dishes. Find a piece of decent looking tissue the size of this (O) and transfer it. Good luck.

Make transfers from that one now.

Did you do multiple tissue pieces to that plate? Your tissue pieces are huge.
Here is a plate from spore syringe using a 1/4 drop.

Gotta start somewhere
I am now only referring to the last pic i posted, which is of the white-only tissue
Like i said, its my first time actually starting my own plates with agar. I tried to use several pieces of tissue per plate. "The pieces in that last pic are pretty small" (i thought at the time, i guess still way too big, but not causing contams that i see) And if my tissue plate looks like "garbage", i would like to know what about it is a problem and particularly if its not a genetic thing. I mean shit, i am just looking to expand into working with agar for the very first time. And if there are reasons that i dont understand why the tissue plate is a problem, id like to know what so in the future i know what not to do. I dont see any contams coming from the tissue plate visually. Maybe im not used enough to agar to notice. But yes i used like 3 of what i thought to be little slices. The pieces in the last pic, i tried to make them really small, but again, first time so i was just experimenting with what i thought was a small amount
PS i dont mind criticism if its constructive.
Anyway though i was gonna make some more plates, and i guess i will try that. I still have the rest of that syringe (and one other PE), could just do that whole .1 of a cc thing and knock up a whole bunch of new ones, and just see where these go. Also will transfer some of the parts that look best to me to a few new plates, and will see what happens!

Transfer to different media and it might help but those do not look vigorous at all. Doing agar will make you more particular regarding the types if growth you consider accptible. Im not even saying it has to be rhyzomorphic. But healthy is important.
Healthy clone

As I'm finding out, agar work and technique varies from person to person. I applaud you for jumping in and giving it a go but it also looks like might have done no research or prep work prior to doing so. Mad has a great Tek Tip in regards to working with agar.

Pasty and Tahoe are great resources as well. If you click on their profiles then click on view previous posts you'll see they have discussed many of your concerns many times.
Otherwise, I'm not sure what else you want to hear.

Well thank you for the tips, thats all i really want to hear. Well, yeah, my friend gave me ingredients (light malt extract) and i wanted to kinda jump into it, because i really wanted to clone the project i had going (and i did) but wasnt able to do as much reading as i wanted to on it because i wanted to clone that flush. Then i figured might as well experiment with some PE plates while im making them. Oh well, like they say on the magic schoolbus... Make mistakes! :P
And maybe something good will come from one of those. I have several more tissue cultures that i havent even looked at, much less taken pics of. Maybe theyre better
I honestly was expecting just like shitloads of contams my first time, so this is way beyond my expectations lol

Yeah, I have been blown away that I haven't fucked up any of the projects I've started. I went into all of this expecting no more than a few tubs of trich, plates of aspergillosis, bags of excess WBS and oats, and kitchen cabinets full of quart mason jars. I guess if you set your bar low... Maybe even place it on the ground, you are bound to surprise yourself. Lol.

Try and see what a transfer does. Might gain a little vigor. Sometimes a transfer helps. But observe the transfer carefully.

I mean there are 3 other tissue plates i havent opened and looked at yet, some might be more vigorous than that one, But what i really wanted to work with was the PE, i was wondering though, if i wanna get some good genetics, does it make more sense to do a regular PE grow with MS and then either g2g or do a tissue clone type thing (i would just use tiny pieces, been trying to find the best links on agar teks besides the ones pointed out, any others would be greatly appreciated, i find lots of confilicting info)....anyway so i could just start ms g2g PE or i could try spores to agar again, i have 6cc left in that syringe, and also a clean PE if need be. Which is the better way to go? Start with spores to agar to ms grow > tissue > agar?
Im also gonna try to look at the other tissue plates and see if any of them are more vigorous, but PE was my main concern (those are TC TC's)

Just start with a drop of PE on a few plates and do an ms grow. Clone from there. 6cc is enough to do 1000 plates.

Thanks, ill try the whole 1 drop per plate thing and see what happens, just in the center. Before i kinda wanted to simulate the sectioned plate thing by dropping spores in diff areas. I'll just do one drop in the middle. Thanks for the advice
