Shroom Syringe
    #2242879 - 01/15/04 12:54 AM (12 years, 10 months ago)

I understand that its possible to start a jar using a already grown mushroom,or part of anyway, however i just read something i never read before. It said that if you inject sterile water into a Mushroom cap right before its vale breaks then suck it back out,you can use this to innoculate jars.Is this true,it sounds very simple, but how sterile can it be?is this a practiced method,i just never heard of it.Are you basicly just sucking out the spores before they drop?

No, you're sucking out fragments of mycellium - this is more like cloning. The idea of this is that the inside of mushroom is more or less free of contaminants, but I never heard of anyone actually trying this and having success.

I found this table that shows that interior of mushrooms isn't sterile.

Quote:

Number of aerobic bacteria found in different basidiomycetes.
Counts are made on King's B agar.

Species M cfu / g F.W. ? S.E. %fluorescent Pseudomonas

Cantharelllus cibarius 2.4 ?1.4 n=35, f=88 78 / 18-98
Forest soil (depth 1-5 cm) 1.8 ?0.92 n=9 12 / 9-22
Sarcodon imbricatus 0.083 ?0.03 n=2, f=4 0
Hydnum repandum 0.30 ?0.03 n=6, f=14 0
H. rufescens 2.5 ?1.1 n=9, f=26 85 / 65-99

Actually it works like a charm. I did this a hella long time ago on a lark, cause I assumed it should work, and it did.

The main issue I had was that you can't inject that much water into a fruit body in the first place.

In the MMGG it shows you how to make a liquid Innoc. with just plain water, and a healthy fresh fruit in a blender. This is the same idea and you get a hell of alot more innoculate out of the deal.

Also you can take the interior of a sliced shroom to grow a clone on agar.

After I build my new glovebox this weekend, I think I may try to slice up a shroom and throw it into WBS QT jars, to see how that works out.

Spore syringes are always moldy to some degree.
But using the PF jar method seems to deprive molds of oxygen and
the mycelium then eats the mold/spore up.

I've had good luck with agar/petri mycelium and sterilized water with jars. Also have put a young shroom in a blender(alcohol washed) and injected it into birdseed jars with great success.

Spore take longer to start taking over than mycelium from a healthy culture.

i disagree, or maybe im misunderstanding what your trying to say.

>using the PF jar method seems to deprive molds of oxygen

i use spore syringes with methods that allow for plenty of air exchange and i dont notice contams appearing anymore frequently then in a sealed jar.

Our task must be to free ourselves from this prison by widening our circles of compassion to embrace all living creatures and the whole of nature in its beauty."

i'm agreeing with amyloid. syringes should NEVER be moldy unless what you mean by moldy is that some cube spores have germinated, I've only seen that once and it was a very old syringe.
As for depriving the mold of oxygen, that's just nonsence. The PF jar is designed to keep out contams not deprive oxygen, though it does to some degree. Cubs, like most any organism, need oxygen to metabolize. Depriving it of oxygen would slow it WAY down, thus giving contams more time to get their claws in there. the more oxygen it has avalible the better your chance that the jar will collonize before contams get a foot hold on your substrate. Once a contam does infect the substrate you are probably fucked. Contamination on casing can be delt with via amputation and H2O2 misting. I always toss anything that contams invetro.
As to collonizing contam molds...? That i gada see.
