Hello,
I looked for teks on this subject but didn't find any. Sorry if there was already something on the subject.
Could I collect spores from a wild mushroom? Example: I find Pleurotus while walking in the forest can I collect his spores without having a 'laboratory' at disposition? Could it be stocked for a few days? (I am going to a place where I know I could find numerous and quality mushroom that is a few days from home and I would like to bring back spores)

You can but spore prints from wild mushrooms are littered with mold spores and bacteria so you would need to separate it from them on agar. You could also take a clone which might be some what easy if you can bring a GB with you. Get a scalpel, a lighter, some 70% iso, some paper towels and an already made up agar dish. Put it all in the glove box and then wipe everything down including the mushroom with the iso and paper towels. Then heat the scalpel until red hot. Cool it on the stem by the cap then cut down long ways. Take a tissue sample from the center lower part and put it to agar. Then pray for a clean culture to whatever deity you believe in haha This works well with oysters but not sure what else. Good luck!

Pleurotus will grow onto cardboard. You can do some ghetto cloning and clean the culture when you get back.

Ah yes totally forgot about cardboard cloning! Throw some stem butts from oyster on soaked cardboard and that shit will go crazy! Some people actually use the cardboard as spawn right to pasteurized straw as well.

Thank you for the ideas guys! Cloning on agar might be difficult in my situation, I'll prefer the method without glove box even though success rate might be lower. I love this cardboard spawn method, thank you very much, very simple, great learning. I'll follow this tek: http://www.shroomery.org/forums/showflat.php/Number/6225388 At a smaller scale, so I can transport it. I'll have to put it in a sealed box in my backpack where it can still get some air exchange. Maybe a Tupperware with a polyfill hole.
I wonder what do you mean by "clean the culture when you get back" Lost geometer. Could I get rid of contamination if there is any and keep only the healthy mycelium? how would I do that?

Once you have mycelium growing on your cardboard you should be able to take samples of the cleaner looking pieces and put them on agar, hopefully giving a clean culture for grain spawn or whatever. The cardboard clone will always be contaminated, even if the contams can't grow well, although I have had some success with wild oyster -> cardboard -> bulk.

I've heard of people colonizing cardboard and then taking a small square of it and dipping it in a 10% bleach solution or diluted iodine (not sure what percentage though) for 5 minutes or so and putting it to agar. Apparently mycelium is quite tolerant to bleach in small concentrations. dumbfounded1600 put together a helpful post of all of RR's notes. Search for it. I learned alot about cleaning up cultures from that. Good luck man and keep us posted!

Yes, good luck with that. I dont think air exchange has to be an issue though. You could probably just put the wet cardboard with the sample in a clean plastic bag and take it home. Then I would say put it in room temperature or a bit warmer if you have a good place, with the bag a bit open for colonization. It might be wise to use polyfill in the opening of the bag when it colonizes, to avoid additional contams.

I think I'm having some luck going from tissue to cardboard to grain with eryngii.

I found and printed a nice oyster I found this spring and used the spore prints to create spore syringes which I used to create grain spawn. Their was some jars that were lost to contamination but I decided to do a large quantity of smaller jars for this reason. Out of 18 jars, 4 were lost to contamination. I believe agar would have been a better choice, still working on that myself. I sort of enjoy the process of sterilizing grain and have had a pretty good success rate using it with living tissue and spores. I believe the jars that were lost to contamination were lost because of a dirty spore print as I generally have a much higher success rate. Choose your specimen carefully.

Thanks to everybody for all these good advices. I will go there in a week or so and I will take pictures so if it works I'll share. In sum: 1- Choose a good specimen, 2- Put it in prepared cardboard 3- Isolate mycelium on Agar once home 4- inoculate grains
