    #22407235 - 10/20/15 05:14 AM (8 years, 3 months ago)

Alright I got a dish with pennicillium that I wanna take a transfer from... how do I go about doing this, should I isolate both dishes together in my SAB? I've never dealt with this contam on agar before...should I just say fuck that dish or what.

Check this out... I didn't strain my rye soak water I was using to make that agar, so in turn there is some floaties in it (it settled for over a week in my fridge)
and I an only PC'ing for 30 minutes when I make my agar, could that be causing a problem?

I dont think so 30 minutes should be plenty are you doing pour or no pour agar? And I put both petris in a sab at the same time for the transfer just try not to disturb the air much on the tam plate l always open the lid slowly and kinda slide it over enough to sneak my tool in the tsm plate to cut a wedge scoop it up and seal it up as smoothly as youbopened it my thought process about this is that the less air gets stirred up the better

That's what I did... just a 1 on 1 transfer in the SAB after my other transfers... it went alright I guess, we shall see... I've never took a transfer from an agar plate with a green mold I usually just toss...I pour my dishes tetherface.

It's a mold contamination so even if you PC for 1 minute it wouldn't be the cause. Anything over 140f for a while would torch the mold.

Wait.

Why did you transfer anything from a petri with green anywhere aside from a different petri?

The mold got in during the inoculation of the dish in question they're satellite colonies not near the transfer. It's from a mess up

word, i always like to be safe

if it looks questionable it'll look much more questionable on a petir dish and a day
