Germination Rates

Hey Everyone,

I am just curious about germination rates. My first batch went 10 out of 12 1/2 pint jars succesful germination. I reused the same jars and my second batch went only 3 out of 12.

Now I have 5 out of 8 1 pint jars. I just inoculated another 12 1/2 pint jars and have not checked them for germination rates. But it is still the same jars from the beginning.

Just curious what percent success you see in general. Do you reuse jars? If a jar fails to spawn, do you reuse the verm/brf mixture or do you dump it?

Just curious. Let me know your experiences.

Nope.  Even if mycellium didn't germinate there is a good chance other things did.  Resterilization messes with the water contents, mold/bacterial metabolites/corpses may not be a friendly place to grow, and the verm barrier may have been disturbed.

It's a pretty bad sign if you aren't seeing germination in a every single jar at every single inoculation point.  Even if you have a dirty syringe, the mush spore count should be massively ahead of anything else and should show some growth at every inoculation point.  I've never had 0 germination and I've used some 0 fucks open air print alcohol sanitized cup syringes.

I'm suspicious of your syringe(s).  If your substrate was the consistency of a gooball, that would cause problems as well.  Even insufficient sterilization wouldn't stifle germination that much.

PF cakes can easily stall from a dirty syringe.  Not sure exactly what to make of your germination rates given I don't quite know how you re-used those BRF jars, but I would expect them to fail most likely.

1 Pints are more likely to stall since beyond the initial plume of mycellium from spore germination, it must creep through the PF substrate, which I believe is rather dense and difficult for it to get through.  The more time it takes to colonize, the more opportunity there is for contaminants to get rolling.  Can't shake to inoculate the whole thing like grains.  Pints+ PF's are more likely to be successful with an excess of Liquid Inoculant/Liquid Culture.  It's rare to see anyone do that, since LI/LC requires skills and equipment that make PF tek irrelevant.  Sprongiform did a bunch of that kinda shit, his grows are p dope.

P.S.  It seems there may be some problems with grain flours other than brown rice if you try to steam sterilize them rather than pressure sterilize.  Also, your pints would require increased sterilization time.

I spawned some popcorn casings and had double-overlay cause I didn't put enough hydrogen peroxide in my automated aquarium mister.  I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave.  I think it will be good cause B+ is so potent.

Maybe your dry verm layer is too thick or you didnt wait long enough after sterilization and killed em with fire.

Well here are my notes on jar prep:

Mixture is 2/3 cup verm : 1/4 cup brf : 1/4 cup water. 3/4" verm barrier on top.
Sterilize with a pc on high for 60 minutes. I have recently read I should only be doing 20 minutes because it may be fucking up the water content.
Wait 24 hours from pc to inoculation.
Needle is flame sterilized between each jar. Perhaps this is my issue. Can there be too much flame? I wait for the needle to turn red, then press very slightly so the needle tip shoots out mist and cools down the syringe.

That's all I can think of in possible places of errors...

You should be safe from gooey consistency with less than 2:1 verm to water.  With 60 minutes in the PC it might be getting dry since you're already using less water than normal.  Take one of your failcakes out and see if it feels too dry to be hospitible.  AFAIK 60 mins is standard, though my cakes shrink and harden quite a bit from that amount of time in the PC, but they seem to work well enough.

Otherwise my bet is on a shitty syringe for such a low germination rate.
