New project questions updated with pics

This weekend I'm going to try pouring my own agar plates. I recently got a 1000ml media bottle and 20 100x15mm petri dishes. The plan is to use 500 ml of water, to 10g of telephone brand agar to 8g of lme. Does that sound right? Throw it in the pc cook it at 15 psi for 15 min. wait for the pc to come back down to atmospheric pressure and agar cooled enough handle. Take it into the sab where dishes and a cup of hot water will be waiting. Pour dishes and wait put water on top, Wait to cool and solidify. Now, I have some myc growing on no pour plates I did a while back. To take a sample from amother plate. I can flame sterilize (red is dead) a utility knife dip it in agar to cool it then take a sample from the outside growth and put it on the new plate?

Nailed it. Once my unused petri cool I put four each in a new quart ziplock. Those I transfer get wrapped in parafilm.

if you take your media bottle and put it in some hot water and wait till everything is about 47-50ÂșC and then pour you'll avoid almost all condensation and avoid the hot water on top of the dish shit.
most people pour their agar while it's too hot since they don't have a water bath, or are worried about all the wrong shit when it comes to sterile technique.
after they cool, wrap the dishes in the SAB and then you can take them out and can incubate them on a bookshelf, no ziploc baggies needed, kind of a waste of baggies in my opinion.
or the same sleeve they came in, it's sterile on the inside, you can just put the sleeve back over the dishes and keep them in there. I keep a sleeve of poured dishes in my fridge just in case I need a petri right away. if you put less than 15 back in the sleeve you can twist the end and fold it under to keep it closed up and nice ready to use again.

The water bath is just to cool the agar before pouring, still have to do the pouring in the SAB, build one that isn't cramped....

Sorry I was stoned when I replied to that. Yes I will be pouring in my sab I was referring to the cup of hot water. I apologize for the confusion.
