Inoculation question

When  sterilizing the jars, I flipped the pins upside down for the sterilization. Should I leave them like that during inoculation and not open them at all till they fully colonize, or I should put them back to normal? Im asking because I want to eliminate any chance of contamination, and opening them might raise the risk.

No. Just the lids are upside down, so the rubber ring faces the sealing.

Is there a cfd or tyvek under those lids? If not there needs to be, or polyfil stuffed in there.

I am more confused than you bro. Im virging on these things. I follow the tek i posted above and thats it. No vermiculate not shit. Just rye. i didnt have the sdf so i didnt use one. I will seal the holes with poly fill after inoculation. If you have any advice give it here.

My advice is to put the poly in before the PC cycle. This will sterilize the filter, and keep it sterile until you inoculate. Do you have a still air box? If you do you can just pop the lid, tilt the jar and drop 3 drops of solution down 1 side. Then put the lid back on :smile:.

Well having open holes in your lids after pc'ing is all bad, they were sterilized but I doubt it now... make up another batch after you get poly stuffed in those holes...pc them with the poly stuffed lids covered in foil.

The jars are still at the PC. I havent opened anything yet. I was sanitizing a bathroom and the shower cabin (Ill inoculste inside the cabin). I will open the PC when im in there with still air box.

I took the cut outs of the holes and taped them to make little doors. This is to keep the air still if I'm waiting for things to cool, or if I want a smoke break.

My SAB holes are 6 inches. Honestly this is the MINIMUM diameter. I took incense and lit it in my box (kudos to kizzle for the idea) and found the arm holes moved the most air (smoke) when moving my arms around, and in and out. By increasing the size of the arm holes, and working in a fairly still room with the vents turned off .. I've been finding that moving your arms in and out will cause no air movement. And thus very rare contams.

I used synthetic filter discs instead of polyfil and have made a couple different types of lids. Some with silicone injection ports, some without. No issues with contaminants yet with either.

Really easy to make, but you do have to plan ahead before PCing your grain.  :shrug:

Thanks for the help guys. I will take the risk got nothing to lose. The bathroom is clean. The shower cabin where I will do the magic was bleached and poured with boiling water x2. No air flow. Finishing the still air box, and getting inside. I will open the PC when I am in there, so the risk of contam is low low. I use surgeon clothes and mas and hat, sanitized twice. Gloves, mask, goggles. I think it should go smoothly and successfully.

When using MS you shouldn't shake after inoculation until 20 to 30% colonization. It's helpful to shoot the solution along the side of the glass so you can see the mycelium form and watch for contaminants. If you end up inoculating the middle of the grain you won't have an idea if the jar is colonizing until the growth is pretty far along because it will work its way from the inside out.

Yeah, thats what Im thinking of. 1 drop  on two sides. I use 12oz jars, pretty small.. but my PC is small and i cant PCing all day...

I'm doing both right now! That and Argentina on agar and Malabar on agar and MS to oats.

They all seem kinda the same to me. By day 4 each time I have had growth in my jars and agar after inoculation.

What a fail ladies and gentleman. I prepared 1:30min for this shit, I opened the PC and saw half of my jars broken, the rest still at 1 piece with burned rye at the bottom.  :rockon:

There was no water left, but it might have been consumed by the rye. And no... I didn't put anything at the bottom. That thought was bugging me the whole time, but I didn't put anything after all. Oh well... Tomorrow.

I'd work on not letting the weight jiggle. Sometimes you gotta tape some coins or something onto the weight, and you can get it to hover at any pressure if you adjust the burner accordingly.

If your PC came with a trivet, use it, if not... Make a layer of jar rings on the bottom of your PC, put a layer of foil over the rings. That makes a little platform so your jars don't touch the bottom. Water is VERY Important, read your manual that came with the PC...mine tells the minimum amount of water for a cycle. Some PC's have a line to mark it for you.

Is it normal when the water starts to boil, to throw some on thentop of the jars. I had thr fear not to put too much water, i was thinking that this might flood the jars somehow :laugh:

I followed the user manual al to my PC regarding how much water should be added, which ended up being 3 quarts of water. Maybe you can look you PC up online and find its user manual so you know how much water to add?

I'd say my jars are a 1/3 of the way submerged when it's full. Don't worry, you'll always be learning something new! I just figured out this week that I could stack my jars in the PC. I thought they had to be submerged in water to safety PC. I don't want to talk about how many times I have ran my PC half full. Talk about inefficient.

Look for a manual online - nothing... I guess the water didnt evaporate fully, but when the jars broke and the grain spilled, it consumed a lot of the water. Thats possible right?

The foil on top of your jars is there to keep water from the lid dripping into your jars throwing off the water content... so you definitely don't want to throw water on top of them on purpose... when I did pf tek I used my steamer pot with a row of lid rings under the rack to keep my jars completely out of the water... bought a pc and never looked back.

It is possible, and probably more than likely, run a test run on your pc and see if it leaks...if it did run dry it will probably be warped and not seal.

The only problem with that is when you flame your needle then stab the poly it melts and sticks to the poly...just inoculate in your sab, Crack the lid and inoculate like that.

I thinkg I did well. My only concern is the PC. I did pretty well with cleaning the room where I'll inoculate. Its in a bathroom in the shower cabin with e still air box. I cleaned so much, I can eat from the floor. :laugh:

Well that's good practice for cleaning your house... but it's really overkill for a SAB, spray the inside with soapy water, have a wet towel on the bottom and be in a room without a draft...sanitize your hands and arms and go to town...you don't need to be in the shower... but hell, high five on the extra precautionary measures.

About the PC it didnt go as some people told me. It didnt whistle couple of time in a minute. It was constantly just making that pressure noise. I haven't opened the PC yet, I fear that the jars might be broken again... Fingers crossed.

Room temp... 75 is optimal, but don't start over thinking this part... if it's not continously freezing, or scorching hot they are fine... that's why you need to get into agar, keep you busy...this hobby is stressful if you don't have multiple projects going... AGAR FTW.

That's not an abbreviation... it is called agar agar... it is a gelatin that we mix nutrients into, pressure cook it, let it cool and set up in petri dishes or pp5 (plastic that has the "5" recycle code on it) dishes...then drop one drop of spore solution on it in your sab, let it grow out, take a scalpel and transfer wedges of mycelium to another agar plate... used to clean up cultures... (I never go spore syringe to grains, too big of a chance of unclean spawn)it is mandatory for great success in this mycology shit.

Believe me... before starting all this i have read every possible pf tek i found. From top to bottom, researched a lot. but i wanted everything to be absolutely perfect, i didnt just want to sterilize bunch of jars with rye and stick a syringe and a needle inside. For me doing it with quick and not in detail is the same as somebody puttig me at e half built house with racoon shit inside. If i want to grow up and develope fast and strong, i need a perf environment. So this goes both ways. I hope you wont think im totaly nuts.

I hear ya. I'm about a month or into my first grow myself. Nothing but good experiences so far but I spent a tremendous amount of time reading long before I decided to start. Like you I've been asking an overwhelming amount of questions. Sometimes just for validation.

I've never done cakes before. I went straight to bulk simply because the cakes seemed... Disappointing. But the more time I spend online talking to folks the more I see why people choose to do them. I might even give them a try one of these days.

Switch to SFD's (synthetic filter discs) when you can, they are sold online. But if you use Polyfil (I did for a long time with great success) You HAVE to make sure to stuff that lid as tight as possible. You should test it by pulling on it from both sides and it should not move without a ton of force. Everyone has a different take, but I used to inoculate straight through the Polyfil. Just heat your needle red hot, then take it into your SAB, squirt out a drop of spore solution... This will cool the needle, Gently insert needle in Polyfil and point toward the side of the jar so the solution runs down and pull out carefully. You should feel resistance when you put your needle in, the Polyfil should be that tight....Anyway Good Vibes and :goodluck:

I had probs with the inoculation tonight. When I heated the needle on a candle, and it got black, and then that black layer got off the needle and I am afraid is in my jars now :| Any advices?!?!?!
