How many transfers should I perform on agar, and how large of a wedge should I use to inoculate?

I don't really feel the need to post any pics, but I put ATL#7 spores on two agar plates, and one had shown signs of growth after 3 weeks. I saw what looked like bactria colonies, so I took two samples of healthy mycelium from the side opposite the contams and transferred to two new plates. Is it necessary to perform a second transfer with each of the plates? How many transfers are necessary before a consistent strain is isolated? Also, how large of a wedge should I inoculate my quart sized oat jars with? I was planning on allowing the mycelium to grow out to roughly 2/3 the diameter of the plate, then use about 1/8th of what is there. Kinda like a pizza slice wedge.

i use as much agar as possible to inoculate.
if u were running from bacteria, that next plate should be fine to use. if ur running from mold, i'd say do another transfer.
if u are trying to isolate from spore, it could take many transfers. if ur just trying to noc up some jars, no need to do that really unless u want to. noc grain with clean MS>bulk>clone
or let plate fruit/form stones>clone from the plate.

Okay, sounds like a plan. Hopefully I won't see any more contams on either plate. I'm really excited for this project since it is my first time working with a sclerotia producing species.

sweet. sclerotia are awesome. post a pic of new xfer plate if ur unsure. but IME, as long as the bacteria is not intertwined with the culture, and just a separate colony on a plate, its easy to get away from.
mold is a harder problem to tackle.
