Cloning Wild Fruits 12

Going to doc a couple ways I like to do this, from typical cloning to wild fruits to dried fruits. To start this I got my sterile water ready, two small ones have a wee bit of bleach
. From there I will either take the two small bottles with me in the field or just use them at home but they are my starting point.
When in the field I simply cut the cap off after pulling the mushroom from the ground, I then wipe any debris off the base and will drop the tissue in the water and shake, this rinses off a lot of contams and the bleach helps as well.
 The next step is removing it once home and putting it in another jar of sterile water and shaking a few times, this step can be repeated a couple times and this greatly waters down any contams present and gives your tissue sample a much better chance and without things like Iso and iodine which may make things harder!
 So we took some gyms and did steps one and two and proceeded to clone like normal taking the last sterile water jar with the tissue sample to the sab for final removal before cloning to no pour Petri's with premixed mea.
 After wiping down the water jars and agar jars in iso and prepping the sab like normal I took the tissue sample out from the water and tore it in half from the top down and put three samples in three agar jars. Within 3 days I had great growth in them and was ready for the first transfer in just four days.


A few transfers in....

.
 ready for grain
 for the first run though i'm going to go to supplemented jars(wheat gypsum coffee bran wodchips)
\  Here i did this with an Agaricus mushroom.
  Cut the stipe
 Add to jar 1 and shake
 Transfer to jar 2 and shake and repeat for about 3 jars of water/bleach
 Then take it out and place it in your prepped sab.
 Tare it from the top down in half
 Take your tissue sample from the inner base
 Place on agar wait for growth.
Here's a quick over view on how i did the dry fruit clone......no different from what you see above otherthen the first soak sits for a bit
 dried fruit

I've seen a couple different posts about cloning wild mushrooms, and I'm having trouble seeing why cloning a wild one would be any different than cloning one from an indoor grow. Yeah, maybe outside is dirtier and more contaminated (although after accidentally seeing the some of the shit that is more than happy to grow on agar in my bedroom, I might challenge that statement), but if the steps for cloning a promising mushroom from your grow (or really, for anything involving contams, ive had to follow this process to get something out of a horribly contaminated syringe) are:
Step 1: Get a bit of interior tissue onto agar Step 2: transfer away from contams, repeat until you have a clean culture
Then why do wild mushrooms require special treatment?
I'm not being at all dismissive here, this is an honest question, because I'm probably going to hunt edible species in my area at some point.

Outdoors is strait up dirty! i can't remember the numbers exactly but trust me it's friggiun insane how many contams can be found inside and outside of a fruit

So, do I sterilize a container and then poor the sterile water into the said sterile container, and let it cool few a few hours before applying the mushroom to the water? Or do I sterilize the water using a cap of bleach and just toss the mycelium into it?

I've got two jars I wanna do invitro. They're fully colonized but smell a tad bit bacterial. Should I birth them into a bleach water rinse and then clean the jars, and put them back into the jars with a vermiculite casing layer on top?

no bacteria is not much of a threat to a fruiting sub, it can defend itself against it at this point just make sure to give it some air if possible

I've got some PE6 that stalled in the jars on my second failed growing attempt. I buried them in my cow poo pile and now they're fruiting. I picked a couple that weren't quite open yet. I think I'm going to try cutting the veil with a sterile knife to get a clean spore syringe, and also try this tek with some tissue culture. I'll have to try an LC instead of agar (no PC). Keeping my fingers crossed and I'll let you all know how my attempt goes.

You may get away with agar with no PC a few times but it'll never be a reliable method.

Sugars tend to carmelize with long term heat, when I did it my agar was very dark

Got pink mold in my one of my unbirthed jars; Considering that it's from bacteria (according to an article I read anyways), think that a bleach water bath would off it? This house is so fucking gross

Well, I'm soaking it in bleach water regardless. The pink appears to have faded significantly in moments. I'll just fruit it outdoors or something.

first transfers of the clitocybes in the morning,took 3 samples all 3 were clean! also got the agaricus to do as well,gyms are going into fruiting tonight

I wonder if instead of trying to boil this bacterial syringe, I can just fill it with bleach water, and leave it full until I'm ready fill said syringe with LC, and/or skip flame sterilization in between agar transfers and just have a shot-glass full of bleach/water in the SAB to dip the syringe in in-between transfers.

Might not be exactly relevant but I got a dry fruit question:
Got a dry cube with a dark red cap from a friend. It was harvested before the veil even began to stretch, when it still has that roughled colar look. Can I crack the cap open and pull some spores with a swab?
Would it be preferable to put the spores into a syringe to rehydrate, rather than swab directly to agar?
It appears to have been grown on a cake, so I'd rather start with vigorous MS growth than a tired clone of a random ms fruit. I'll try both methods though.

I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave. I think it will be good cause B+ is so potent. Triggered yet? Only a square would say "a cube is a cube."

you can try to swab it for sure, if there are mature spores it will work fine, no need to rehydrate agar is great at thatbas well.

I hadn't even considered spore maturation. I assume they would be mostly mature. I'd place this mushroom's age as about 1-1.5 days before veilpop.

If I do clones of something with a meaty cap I prefer that. It tends to cut out nicely.

Exactly what i needed to read a couple of days ago!
Put grossery oysters to water agar, something that looks like bacteria all around and no mycelium stretching out.
This tech comming up next!
I have a project ressearching antibacterial properties of some plants in agar. Guess this would be the perfect application to try my hypothesis on.
