Hello all! Knocked up 18 jars in a simple SAB about a week ago following RR's instructions. Half are GT half are Amazon. So far the Golden Teachers are taking off but no signs of life in the Amazon jars. Here are a couple pics of some of the the GT jars at 6 days. If these are successful I plan to take some live tissue cultures to agar and go monotub.

Will update in a few!

Any idea why the Amazons aren't doing anything? All were inoculated at the same time under the same conditions. I used a ms syringe. Any thoughts?

Could be a bunk syringe. Could be you may not have gotten spores through, but doing that many jars seems unlikely that you would have missed all of them. Id give the syringe another try though before singling the syringe out as the problem especially if you got them from a trustworthy source. Maybe try in an lc . sometimes the genetics just take a while.

~1cc per jar. Spores were visible to the naked eye in the syringe. I'll give them another week, hopefully they come around, if not I'll just toss them and try again.

They both work from reading it's that the wide mouth colonizes a bit better because it's shorter and also it's easier to wiggle out the cake

Yes the skinny ones CAN work BUT they have a tendency to stall out because air doesn't flow well to the bottom giving more time for contams to find a way to ruin your cake.

12 days in, still no activity from the amazons, I'm going with bunk syringe. Better luck next time.
I'm going to try re-sterilizing the 9 jars and knocking them up with another strain. Any suggestions?

I was actually looking at both of those and plan to grow them sometime. Albino A+ and penis envy also caught my attention.

That's my plan, but since the jars are gonna be well over 2 weeks old by the time I get another syringe I'm thinking about shooting a couple ccs of water in each jar and sterilizing them again before inoculation to reduce the chance of contams/add water that may or may not have been lost by the jars just sitting there.
But for the record you should just make fresh jars. Brf cakes are super cheap/easy to make. I just want to see if they'll colonize.
